摘要
目的建立活血液的质量标准。方法采用薄层色谱法对活血液中的当归、川芎、延胡索3味药材进行定性鉴别;建立高效液相色谱法(HPLC)测定羟基红花黄色素A含量的方法,色谱柱为Diamonsil C18(150 mm×4.6 mm,5μm);流动相:甲醇-0.4%磷酸溶液(30︰70);流速:1.0 ml/min;检测波长为403 nm;柱温:25℃。结果在薄层色谱中均能检测出当归、川芎及延胡索的成分,阴性样品溶液无干扰;HPLC法测定羟基红花黄色素A可达到基线分离,进样浓度在5.06~50.60μg/ml范围内质量浓度与峰面积呈良好的线性关系;羟基红花黄色素A的平均回收率为99.67%(RSD=1.05%)。结论本方法简便、灵敏、重复性好,可用于活血液的质量控制。
Objective To establish the quality standards of Huoxue Liquid. Methods The qualifications of three herbal plants of the Huoxue Liquid: Angelica sinensis, Chuanxiong Rhizoma and eorydalis turtschaninovii were de- tected by thin-layer chromatography (TLC) ; content of hydroxyl saffloryellow-A was detected by the high - pressure liq- uid chromatography (HPLC) on a Diamonsil C18 chromatographic column( 150 mm × 4.6 ram, 5 μm) , the mobile phase was methanol-0.4% phosphoric acid solution (30 : 70) ; the flow rate was 1.0 ml/min, the detection wavelengths was 403 nm, and the column temperature was kept at 25℃. Results Contents of the three herbal plants were detected by TLC without interference of negative samples solution; hydroxyl saffloryellow-A could achieve baseline separation, and the better linear relationship between mass concentration and peak area was achieved with 5.06 - 50.60 μg/ml of sample in- troduction concentration; the average recovery was 99.67% ( RSD = 1.05% ). Conclusion The method is simple and ingenious with better repeatability, and can be used to control the quality of Huoxue Liquid.
