植物源化合物百里香酚对西瓜枯萎病菌的抑菌机制初探 被引量:14

摘要 为了明确植物源化合物百里香酚对西瓜枯萎病菌(Fusarium oxyporum f.sp.niveum)的抑菌活性及机制,采用室内生化测定方法研究了其对西瓜枯萎病菌菌丝生长量、分生孢子产生及萌发、细胞膜通透性、蛋白质及DNA合成的影响,并通过显微镜观察菌丝形态的变化。结果表明,百里香酚对菌丝生长量、分生孢子产生和萌发具有较强的抑制作用,其EC50值分别为42.91 mg/L、35.18 mg/L和56.46 mg/L。经百里香酚处理后的菌丝生长缓慢,细弱,分支减少,分生孢子的产生受到抑制;百里香酚浓度为25 mg/L时,能够显著影响菌丝体细胞膜的电导率,电导率随着处理时间的延长上升明显。浓度为12.5 mg/L时,百里香酚对西瓜枯萎病菌菌丝体的蛋白质合成有一定的抑制作用;浓度为100.0 mg/L时,其对菌丝体DNA的合成有明显的抑制作用。说明百里香酚通过改变西瓜枯萎病菌菌丝体细胞膜通透性、抑制菌体内部分蛋白质及DNA的生物合成达到抑菌效果。 This experiment aimed to reveal the antifungal activity and inhibitory mechanism of botanical substance thymol at different concentrations to Fusarium oxyporum f. sp. niveum ( FON ). The effects of thymol on the mycelium growth, conidiophore production and germination, myeelium membrane permeability, protein biosynthesis and DNA biosyn- thesis of FON were studied by indoor bioassay method, and the changes of mycelium morphology through a microscope were observed. The results showed that the toxicity of thymol was stron~ in controlling FON. Thvmol could intensively inhibit themycelium growth, conidiophore production and germination of FON, and the ECso values were 42. 91 mg/L, 35.18 mg/L and 56.46 mg/L, respectively. Microscopic ob- served that the mycelium branch and conidiophore were fe- wer than that of CK. Thymol was found to influence the permeability of the cell membrane of FON at 25.0 mg/L, and the conductivity increased significantly over time.Thymol inhibited protein biosynthesis mycelium of FON at 12. 5 mg/L and DNA synthesis at 100.0 mg/L. The antifugal activity of thymol against FON was exerted through changing the permeability of cell membrane, inhibition of the biosynthe- sis of DNA and protein.
