正交设计和动态过程优化两面针半仿生提取工艺

摘要 目的优选两面针的半仿生提取工艺条件。方法以总生物碱得率和干膏收率为指标,采用正交实验优选半仿生提取工艺后,再动态过程精选提取工艺。结果最佳工艺条件为:复合酶预处理后再70℃水浴提取,第1次以6倍量pH 2.0缓冲液提取80 min,第2次以6倍量pH 7.5缓冲液提取60 min,最后以6倍量pH 8.3缓冲液提取50 min。结论该工艺高效、节能、省时,为工业生产奠定实验基础。 Objective To optimize the condition of semi-biotic extraction technology for Zanthoxylum nitidum. Methods The extraction technology was optimized by orthogonal test followed by dynamic process, with extraction rate of total alkaloids and dried paste weight as the indexes. Results The optimal process was as follows: after pretreatment with cellulase- pectinase,Zanthoxylum nitidum was extracted in the 70 ℃ water bath for 80 min with 6 times volume of pH 2.0 buffer for the first time,then for 60 rain with 6 times volume of pH 7.5 buffer for the second time,then for 50 min with 6 times volume of pH 8.3 buffer for the last time. Conclusion The process is efficient, energy- and time-saving. It establishes an experimental basis for industrial production.
