摘要
采用剪碎法对从广东省湛江市某肉猪屠宰场随机采集的100多份肺组织样品进行猪肺炎支原体野毒株的分离,结果分离到1株疑似猪肺炎支原体;通过培养特性、形态观察、生化试验、血清学试验初步鉴定其为猪肺炎支原体并命名为GDZJ01。用PCR方法扩增分离株P46基因、P65蛋白C末端的基因编码序列和P97蛋白R1R2区域的基因编码序列,并测定其核苷酸序列。结果表明,P46基因和P65基因片段高度保守,与标准株J株、232株和7448株的核苷酸序列同源性分别为98.9%~99.1%和99.2%~99.7%,而P97基因片段的变异较大,相应地,核苷酸序列的同源性为86.6%~96.0%。这些结果进一步证实分离株GDzJ01是猪肺炎支原体。
A suspected Mycoplasrna hyopneumoniae was isolated from more than hundred lung tissue samples randomly collected from a slaughterhouse of Zhanjiang Prefecture of Guangdong Province by the method of shearing. According to the results of morphological observation,culturing and biochemical characteristics detection and serological test,the isolate was tentatively identified as Mycoplasrna hyopneurnoniae and named GDZJ01. With PCR method,the full-length P46 gene and partial sequences encoding P65 Cterminal and RIR2 domain of P97 were amplified, respectively, and sequenced. In result, both of P46 and P65 genes were highly conserved and the nucleotide homology between GDZJ01 and the international standard Mycoplasrna hyopneurnoniae strains,J,232 and 7448 was from 98.9% to 99.1% and from 99.2% to 99.7% respectively,whereas the R1R2 domain of P97 gene fragment showed relatively higer variation, and the nucleotide homology between GDZJ01 and the standard strains was from 86.6% to 96.0%. The above-mentioned results demonstrate that GDZJ01 is Mycoplasma hyopneumoniae.
