泡桐叶绿体游离及其DNA提取方法的筛选

摘要 为了深入研究泡桐叶绿体基因组以白花泡桐组织培养苗为材料,利用蔗糖密度梯度离心法、差速离心法和高盐低pH值法3种叶绿体游离方法提取叶绿体DNA,其质量浓度分别为92.1、968.9、175.9ng/μL。经显微镜观察、凝胶琼脂糖电泳检测,结果表明,先差速离心提取粗叶绿体,再用蔗糖密度梯度法对其进行纯化,然后使用SDS法提取所得的叶绿体DNA纯度较高,此方法可用于叶绿体基因组的研究。 To further study the Paulownia chloroplast genome, the Paulownia fortunei seedlings derived from tissue culture were used as test material to extract the chloroplast DNA by three methods, sucrose density gradient centrifugation, differential centrifugation and high-salt low-pH method, the mass concentration of which was 92.1 ng/μL, 968.9 ng/μL and 175.9 ng/μL respectively. The results of microscopic observation and agar gel eleetrophoresis indicated that the best method to obtain the pure cpDNA was to extract the gross chloroplast firstly by differential centrifugation, then purify the gross chloroplast by sucrose density gradient centrifugation, and finally get pure cpDNA by SDS method, which could meet the requirements of chloroplast genome research.
