3种植物生长调节剂对培养基中坛紫菜叶状体组织块生长的影响 被引量:2

摘要 为了探讨植物生长调节剂对紫菜组织培养的影响,以坛紫菜(Porphyra haitanensis)叶状体为材料,在培养基中分别加入3种不同浓度的植物生长调节剂(α-萘乙酸、6-苄氨基嘌呤和激动素),观察组织块在生长过程中对植物生长调节剂的反应.结果表明,在培养基中添加1.5~2.0 mg/dm3 α-萘乙酸能够促进坛紫菜叶状体组织块生长,在培养14 d时1.5和2.0 mg/dm3 Q-萘乙酸处理组织块面积分别是对照的1.76和1.58倍.在培养基中添加6-苄氨基嘌呤2~6 mg/dm3培养7d时组织块面积显著地低于对照,8 mg/dm3处理组织块全部死亡,培养14 d时6 mg/dm3处理的组织块也全部死亡,表现出6-苄氨基嘌呤对组织块生长的抑制作用.在培养14和21 d时,激动素浓度为0.5 ~ 1.0 mg/dm3对坛紫菜组织块生长有抑制作用;添加5.0 mg/dm3或10.0 mg/dm3激动素培养7d时,组织块细胞内无色素体结构,全部死亡.试验还观察到,在培养基中添加0.5 mg/dm3激动素时,坛紫菜组织块产生类似于单孢子的细胞. The effects of 3 different plant growth regulators (α-naphthylacetic acid,6-benzyl aminopurine and kinetin) with different concentrations on thallus tissue blocks of Porphyra haitanensis in the process of growth were investigated for the effects of plant growth regulators on tissue culture.The results showed that NAA in concentration of 1.5 ~ 2.0 mg/dm3 could facilitate the growth of thallus tissue blocks obviously.The areas of tissue blocks treated with1.5 mg/dm3 and 2.0 mg/dm3 of NNA were 1.76 and 1.58 times larger than the controls after 14 d culture,respectively.When 2 ~ 6 mg/dm3 6-BA was added into the media and cultured for 7d,the areas of tissue blocks reduced significantly compared with the controls.The tissue blocks would not survive when they were cultured in the media containing 8 mg/dm3 6-BA for 7 d or 6 mg/dm3 6-BA for 14 d indicating that 6-BA could inhibit the growth of tissue blocks.The growth of tissue blocks of P.haitanensis could also be inhibited after cultured for 14 and 21 d when 0.5 ~ 1.0 mg/dm3 KT was added.No pigment structures could be observed and no cell would survive after the tissue blocks were cultured in the media containing 5.0 mg/dm3 or 10.0 mg/dm3 KT for 7 d.Furthermore,it was observed that 0.5 mg/dm3 KT could induce monospores-like cell from the tissue blocks of P.haitanensis.
