Protective Effects of Bone Marrow Mesenchymal Stem Cells on Tubular Epithelial Cells and the Possible Mechanism

摘要 目的探讨骨髓间充质干细胞(bMSC)移植对大鼠UUO模型肾小管上皮细胞超微结构的改善作用及其可能的作用机制。方法体外分离培养雄性Wistar大鼠bMSC,体外Brdu标记,将Brdu-bMSC注入同种大鼠体内。45只受体大鼠随机分为3组:假手术对照组(SH组,n=15)、Brdu-bMSC+UUO组(BM组,n=15)、生理盐水+UUO组(NS组,n=15)。分别在3 d、7 d和14 d处死大鼠,取下肾脏组织,电镜观察肾小管上皮细胞微结构变化。采用免疫组织化学方法检测Brdu阳性bMSC。结果电镜下,随着结扎时间延长肾小管上皮细胞微结构破坏逐渐加重,术后3 d、7 d BMSC组肾小管上皮细胞超微结构明显改善;未在大鼠肾脏组织中发现Brdu阳性细胞。结论 bMSC移植可短期内改善肾小管上皮细胞超微结构,对肾小管间质病变起到保护作用,其作用机制可能是通过旁分泌作用。 Objective To investigate protective effects of bone marrow mesenchymal stem cells on tubular epithelial cells ul-trastructure in rats with UUO, and explore the possible mechanism. Methods BMSC were isolated from male Wistar rats and were marked with Brdu, then injected intravenously at the same time into Wiatar rats with UUO. 24 male recipient Wistar rats were divided randomly into three groups( n = 45 ) :sham operation group( Group SH, n = 15 ), Brdu-bMSC injection group( Group BM ,n = 15 ) and normal sodium infusion group( Group NS,n = 15 ). The kidneys were harversted at 3 d,7 d and 14 d after the operation. The renal tubular epithelial cell uhrastructure was examined by transmission electron microscopy. The Brdu positive bMSC in kidney were examined by immunohistochemical technique. Results With the ligatured time extension, the renal tubular epithelial cell uhrastructure were more damaged. Transplantasion of bMSC could improve the damaged ultrastructure of the renal tubular epithelial cells at 3d,and 7d after the operation. There was no significant deviation at 14 days. There was no sign of la-beled cells residing in the damaged kidney. Conclusion Transplantasion of bMSC could improve the damaged uhrastructure of the renal tubular epithelial ceils in short time, and protect tubulointerstitial lesions. The the possible mechanism may be paracrine effects of bMSC.
