摘要
目的:观察中药提取物三氧化二砷(Arsenic Trioxide,As2O3)对海马神经元Kv4.2、Kv4.3钾通道表达的影响。方法:将SD大鼠随机分为5组,分别经侧脑室注射生理盐水、不同剂量的As2O3(1mg/mL,0.1mg/mL,0.01mg/mL)和4-氨基吡啶(4-AP,15mmol/L),观察给药后大鼠行为学的改变及Western-blot法检测海马神经元Kv4.2、Kv4.3钾通道的表达。结果:As2O3随浓度增加,能够剂量依赖性的引起海马神经元Kv4.2、Kv4.3蛋白表达增强,1mg/mL和0.1mg/mL的As2O3可引起大鼠癫痫样的抽搐发作。结论:As2O3能够影响海马神经元Kv4.2、Kv4.3钾通道的表达并引起癫痫样发作的行为学的改变。
Objective:To observe the effect of arsenic trioxide (As2O3) on expression of Kv4.2 and Kv4.3 of hippocampus neurons.Methods:The SD rats were divided into five groups randomly,and they were given normal saline,As2 O3 (1 mg/mL,0.1 mg/mL,0.01 mg/mL) and 4-amion pyridine (4-AP,15mmol/L),respectively.Then the behavioral changes were observed in the rats and expressions of Kv4.2 and Kv4.3 of the hippocampus neurons were observed by western-blot.Results:As2O3 enhanced the expression of Kv4.2 and Kv4.3 of the hippocampus neurons dose dependently with the increase of concentration.As2O3 induced epileptiform seizures of the rats at concentrations of lmg/mL and 0.lmg/mL.Conclusion:As2O3 could affect the expressions of Kv4.2 and Kv4.3 of the hippocampus neurons and induce epileptiform behavioral changes.
