摘要
目的 :旨在探讨妊娠晚期血小板的功能变化 ,以了解妊娠期前栓状态的病理生理基础 ,为临床诊断及防治监测提供准确、敏感及特异的参数。方法 :应用抗人活化血小板表面 α-颗粒膜蛋白 ( GMP- 1 40 )单克隆抗体 SZ- 51 ,采用放射免疫方法检测 41例孕妇血小板表面 GMP- 1 40分子数 ;用酶联免疫法检测血浆中 v WF水平。 34例健康女性为对照组。结果 :1孕妇组 G血小板表面 GMP- 1 40表达及血浆 v WF水平均明显高于对照组 ( P<0 .0 1 ) ;2孕妇组血小板计数与对照组无差异 ( P>0 .0 5) ;3GMP- 1 40表达及v WF水平间无相关性 ( r=0 .0 2 4 ,P>0 .0 5)。结论 :血小板表面 GMP- 1 40有明显高表达 ,证明孕晚期血小板明显活化 ;但无明显破坏 ,提示血小板功能改变尤为重要。v WF水平增高亦从另一侧面证明血小板活化 ,但不如 GMP- 1 40检测敏感、特异。证实血小板活化为孕晚期前栓状态的重要基础。 GMP- 1 40单抗检测法敏感、特异。
Objective The aim of this atudy is for understanding the functional change of platelets and the patho physiologic basis of prothrombotic state in late pregnancy, and providing a sensitive,spesific parameters for the clinical diagnosis and treatment.Methods Using the monoclonal antibody SZ 51 to recognizes an α GMP 140 on the surface of activated platelets by radioimmunoassay,and investigate the level of vWF in plasma by ELISA in 41 late pregnant women.The 34 healthy unpregnant women were the control group.Results (1) The GMP 140 molecular numbers on the platelets surface and the level of vWF in plasma of pregnant women were higher than the control group significantly( P <0 01);(2)The mumber of platelets was no differance in both groups ( P >0 05);(3) There is no correlation between the express of GMP 140 and vWF level( r =0 024, P >0 05).Conclusion The high expression of α GMP 140 on the platelets surface and high level of vWF showed that the platelets were highly activated in late pregnancy.It was the important patho physiologic basis in the prothrombotic state.GMP 140 McAb assay has the sensitive and specific parameter for platelets activation and it possesses a practical value in clinical.
