摘要
建立了毛细管电泳法快速测定柿蒂中熊果酸和齐墩果酸的方法。考察了缓冲溶液、β-环糊精、有机溶剂添加剂、工作电压以及进样时间等因素对分离检测的影响。在最佳条件下,6.0min内即可实现二者的良好分离,熊果酸和齐墩果酸的峰高和浓度分别在50~1000μg/mL(r=0.9986),25~500μg/mL(r=0.9995)范围内呈良好线性关系。该方法应用于索氏提取柿蒂中三萜酸的测定,回收率在98.36%~104.82%,结果令人满意。
A rapid method for the determination of ursolic acid and oleanolic acid in Kaki Calyx by capillary electrophoresis was described. The key factors such as the effects of running buffer, β-cyclodextrin, or ganic additive, working voltage and injection time were discussed in detail. Ursolic acid and oleanolic acid could be separated and detected within 6.0 min under the optimum separation conditions. A linear response between the peak height and concentration was obtained in the range of 50 - 1000μg/mL (r = 0. 9986) for ursolic acid and 25 - 500 /μg/mL (r = 0. 9995) for oleanolic acid. The method was used to detect the triterpenoidic acid extracted by Soxhlet extraction in Kaki Calyx. The assay results were satisfactory with recoveries in the range between 98.36% and 104.82%.
