Expressions of survivin and minichromosome maintenance protein 2 in breast carcinoma and their clinical significance

摘要 目的检测存活素(survivin)、微小染色体维持蛋白2(MCM2)在乳腺癌组织中的表达,探讨其在乳腺癌发生过程中的临床意义。方法应用免疫组织化学SP法检测内蒙古医科大学附属医院2003年1月至2007年6月收治的60例乳腺癌(全部为浸润性导管癌)及40例乳腺纤维腺瘤组织中Survivin与MCM2的表达情况,数据分析采用χ2检验及Spearman等级相关分析。结果 Survivin在乳腺癌组织中的阳性表达率高于乳腺纤维腺瘤组织,两者差异有统计学意义(12.50%vs 71.67%;χ2=33.661,P=0.000)。Survivin蛋白的表达与乳腺癌组织学分级有关(χ2=9.941,P=0.007)。MCM2蛋白在乳腺癌组织中的阳性表达率高于乳腺纤维腺瘤,两者差异有统计学意义(10.00%vs 58.33%;χ2=23.567,P=0.000)。MCM2蛋白的表达与肿瘤直径、组织学分级、临床分期及腋窝淋巴结转移有关(χ2=4.239,9.307,5.126,6.655;P<0.05)。Survivin与MCM2在乳腺癌组织中的表达呈正相关(r=0.294,P=0.023)。结论 Survivin和MCM2可能与乳腺癌的发生、发展有一定的关系,其表达情况对乳腺癌的诊断及预后判断有一定的临床意义。 Objective To study the expressions of survivin and minichromosome maintenance protein 2 (MCM2) in breast cancer tissue,and explore their clinical significance in the development of breast cancer.Methods The expressions of survivin and MCM2 were detected using immunohistochemical staining SP method in 60 cases of breast cancer (the pathologic type was infiltrating ductal carcinoma) and 40 cases of breast fibroadenoma treated in Affiliated Hospital of Inner Mongolia Medical College from January 2003 to January 2007.x2 test and Spearman rank correlation analysis were used for data analysis.Results The positive expression rate of survivin in breast cancer tissue was significantly higher than that in the fibroadenoma (12.50%vs 71.67% ;x2 =33.661,P=0.000).The survivin expression showed a significant correlation with the different histological grade of breast cancer (10.00% vs 58.33% ; x2 =9.941,P =0.007).The positive expression rate of MCM2 in breast cancer tissue was significantly higher than that in the fibroadenoma (x2 =23.567,P =0.000).The expression of MCM2 showed a significant correlation with tumor diameter,histological grade,clinical stage,axillary lymph node metastasis (x2=4.239,9.307,5.126,6.655;P<0.05).The expressions of survivin and MCM2 in breast cancer were positively correlated (r =0.294,P =0.023).Conclusion The expressions of survivin and MCM2 might be related to the development of breast cancer,which might be helpful in the diagnosis and prognosis of breast cancer.
