摘要
目的:通过检测TLR4在肾阳虚大鼠肾内的表达,为探讨右归胶囊在治疗肾阳虚中可能的免疫学机制奠定基础。方法:健康雄性SD大鼠随机分为对照组、模型组、干预组,模型组和干预组肌肉注射氢化可的松建立肾阳虚大鼠模型,对照组和模型组给予生理盐水灌胃,干预组给予右归胶囊混悬液灌胃。大鼠肾组织常规切片,免疫组化法检测TLR4的在肾脏的表达,Real-time PCR法测定TLR4mRNA的表达。结果:模型组和干预组TLR4蛋白和基因的表达较对照组明显减少,差异有统计学意义(P<0.05);干预组和模型组比较,干预组TLR4的蛋白和基因的表达明显高于模型组,差异有统计学意义(P<0.05);结论:右归胶囊可能通过调节TLR4的表达,影响机体的免疫功能从而发挥改善机体肾阳虚状态的作用。
Objective：By detecting the expression of TLR4 in the kidney of kidney Yang deficiency rats, to study the immunological mechanisms foundation of Yougui capsules in the treatment of Yang deficiency of kidney. Methods： Healthy male SD rats were randomly divided into control group, model group, the intervention group, model group and intervention group by muscle injection of hydrocortisone to establish the rat model with kidney Yang deficiency, and the control group and model group were given saline lavage, intervention group was given the Yougui capsule suspension liq- uid lavage. Rat renal tissue paraffin was for detecting the TLR4 expression in the kidney by immunohistochemical meth- od, and the TLR4mRNA expression was detected by Real -time PCR method. Results ：The model group and intervention group TLR4 protein and gene expression were decreased significantly and the difference was statistically significant （ P 〈0.05 ）. Intervention group compared with model group, TLR4 protein and gene expression in the intervention group were obviously higher than that of model group and the difference was statistically significant （P 〈 0. 05 ）. Conclusion ： Yougui capsule may through regulating the expression of TLR4, affect the body＇s immune function to play the role of improving the body state of kidney Yang deficiency.
