摘要
目的:观察益智仁盐炙前后对肾阳虚多尿模型大鼠肾脏AQP-2与AVPR-V2 mRNA和蛋白表达的影响。方法:通过实时定量PCR(RT-PCR),研究AQP-2(AQP-2)mRNA和加压素二型受体(AVPR-V2)mRNA表达的影响;通过蛋白印迹法(Western blot),研究益智仁盐炙前后对肾阳虚多尿模型大鼠肾脏AQP-2及AVPR-V2蛋白表达的影响。结果:肾阳虚多尿模型大鼠各给药组AQP-2 mRNA和AVPR-V2 mRNA表达倍数(QR)较模型组明显增大(P<0.05),盐炙品组较之生品组AQP-2 mRNA和AVPR-V2 mRNA QR具有显著性差异(P<0.05),各给药组AQP-2、AVPR-V2蛋白表达灰度值也有明显增高(P<0.05)。结论:益智仁生品与盐炙品对腺嘌呤所致肾阳虚多尿模型mRNA和蛋白表达均具较好的改善作用,且盐炙后作用明显增强。
OBJECTIVE To study the influence of Fructus Alpiniae oxyphyllae(FAO)before and after salt processing on the expression of AQP-2, AVPR-V2 mRNA and AQP-2, AVPR-V2 protein in the kidney of Yang deficiency with polyuria model rats. METHODS The effects of FAO before and after salt processing on the expression of AQP-2 mRNA and AVPR-V2 mR- NA were studied by using RT-PCR. Western blot was selected to detect the expression of AQP-2 and AVPR-V2 protein. RE- SULTS Compared with the model group, each treatment group could significantly affect the expression of AQP-2 mRNA andAVPR-V2 rnRNA (P〈0. 1)5), and after salt processing the effects of FAO were strengthened obviously in comparison with the groups of raw materials of FAO (P〈0.05) . Compared with the model group,the groups of raw and salt processed FAO both increased the ratios of gray value between AQP-2 with β-actin and between protein expression of AVPR-V2 with 13-actin(P〈 0. 05). CONCLUSION FAO before and after salt processing had better improving effects on protein expression of AQP-2 and AVPR-V2 of model rats of the adenine-induced kidney of Yang deficiency with polyuria, and the effects were strengthened much significantly after salt processing.
