壳聚糖-海藻酸钠-羟基磷灰石骨支架材料的制备及生物相容性

Preparation of chitosan/alginate/hydroxyapatite composites scaffold and its biocompatibility

摘要 目的 制备一种新型的人工骨支架材料,并探讨其生物相容性.方法 采用溶液共混法制备了壳聚糖-海藻酸钠-羟基磷灰石骨(CS/ALG/n-HAP)复合生物支架材料.将兔股关节软骨细胞接种于复合支架在96孔板中培养3d后,噻唑蓝(MTT)比色法测定软骨细胞的增殖.结果 软骨细胞活性与CS/ALG/n-HAP复合材料中n-HAP含量有关,当CS/30% ALG复合材料中含3%n-HAP最适合软骨细胞生长,细胞生长相对百分率为63.45%;5% n-HAP的CS/30% ALG表面颗粒增多,细胞相对增殖率降至38.39%.结论 CS/ALG/n-HAP复合支架材料无毒,生物相容性好,有良好的研究应用前景. Objective To prepare a kind of novel artificial scaffold and study its biocopatibility.Methods The chitosan/alginate/hydroxyapatite (CS/ALG/n-HAP) composites scaffold was prepared by using solution blending.Rabbit hip chondrocytes were seeded on the scaffolds for compound culture.Cellscaffold compounds were cultured in 96-well plates for 3 days,and then chondrocyte proliferation was determinded by methyl thiazol tetrazolium (MTT) assay.Results The proliferation reactivity of the chondrocytes was related to the content of n-HAP in CS/ALG/n-HAP composites scaffold.CS/30% ALG/n-HAP composites scaffold containing 3% n-HAP was ideal for the growth of the chondrocytes with the proliferation rate of the chondrocytes being 63.45%.However,CS/30% ALG/5% n-HAP composites scaffold had more particles on the surface and the proliferation rate of the chondrocytes was lowered to 38.39%.Conclusion CS/ALG/n-HAP artificial scaffold is cell non-toxic and has good biocompatibility.
