摘要
目的 探讨RNASET2在胰腺癌中的表达及意义.方法 采用逆转录-聚合酶链反应(RT-PCR)法检测胰腺癌(32例)、癌旁正常组织(24例)、人胰腺癌细胞株(3株)和正常导管上皮细胞株中RNASET2的表达,采用Western blot法和免疫组织化学(IHC)方法分别检测胰腺癌冰冻组织(8例)和石蜡包埋组织(10例)中RNASET2的表达.结果 RNASET2在胰腺癌表达明显下降(6.516 ±3.033,P<0.05),在人胰腺癌细胞株BxPC-3、PANC-1和SW1990中表达明显下降(2.814±0.325、2.342±0.454和5.473±0.682,P<0.01);胰腺癌组织和细胞株中RNASET2蛋白质表达相对下降;胰腺癌神经侵犯者癌组织中RNASET2表达更低(5.021±3.102,P<0.05).结论 RNASET2在胰腺癌组织和细胞株中表达下降,可能与胰腺癌发生和神经侵犯相关.
Objective To investigate the expression of RNASET2 in pancreatic cancer and its sig- nificance. Methods The expression levels of RNASET2 mRNA and in pancreatic cancer (32 cases), ad- jacent benign tissue (24 cases), 3 human pancreatic cancer cell lines and benign ductal epithelial cell line were quantitated by using reverse transcriptase-polymerase chain reaction (RT-PCR). The protein expres- sion of RNASET2 was evaluated by Western blotting in 8 fresh frozen tissues and immunohistochemistry (IHC) in 10 paraffin-embedded specimens. Results RNASET2 was down-regulated significantly in pan- creatic cancer (6. 516 ±3. 033,P 〈0. 05) and cell lines BxPC-3, PANC-1 and SW1990 (2. 814±0. 325, 2. 342 ± 0. 454 and 5. 473± 0. 682,P 〈 0. 01 ) respectively. The protein expression of RNASET2 was obvi- ously down-regulated in pancreatic cancer tissues and tumor cell lines. The group with perineural invasion showed lower expression of RNASET2 (5. 021±3. 102,P 〈 0. 05 ). Conclusion The expression of RNA- SET2 was down-regulated in pancreatic ductal adenocarcinoma and tumor cell lines, and this gene might be correlated to tumorigenesis and ~erineural invasion of pancreatic cancer.
