摘要
目的观察宫颈癌介入化疗前后组织中核转录因子-κB(nuclear factor kappa-B,NF-κB)和MMP-9(matrix metalloproteinase,MMP-9)的表达及临床意义。方法采用免疫组织化学SP法检测80例宫颈癌(CSES)行动脉介入化疗前后、70例子宫颈上皮内瘤样变(CIN)及50例正常子宫颈组织(NCE)NF-κB和MMP-9的表达,分析其与临床病理特征的关系,探讨二者在介入化疗前后的表达情况及与介入化疗效果的关系。结果在CSES、CIN及NCE中,NF-κB、MMP-9蛋白阳性表达率分别为72.5%、18.6%、0%和68.8%、15.7%、0%,差异具有统计学意义(P<0.05)。NF-κB、MMP-9表达与宫颈癌不同的病理分级、临床分期和淋巴结转移状况有关(P<0.05),与患者年龄和组织学类型无关,且二者表达呈明显正相关(r=0.872,P<0.05)。介入化疗总有效率为81.3%,且与治疗前比较NF-κB、MMP-9蛋白表达均显著降低。结论宫颈癌患者存在NF-κB和MMP-9的过表达,介入化疗后二者表达显著降低。介入化疗可通过抑制肿瘤细胞增殖、浸润和转移发挥治疗宫颈癌的作用。
Objective To investigate the expression and significance of NF-κB and MMP-9 in cervical carcinoma tissues before and after interventional chemotherapy ( IC).Methods The expression levels of NF-κB and MMP-9 were detected by immunohistochemistry method in 80 cases of cervical carcinoma ( CSES) before and after IC,70 cases of cervical intraepithelial neoplasm (CIN) and 50 cases of normal cervical tissues (NCE).The therapeutic effect of IC and the correlation between the expressions and clinical pathological characteristics before and after IC were observed and analyzed .Results Among CSES,CIN and NCE tissues,the positive expression rates of NF-κB and MMP-9 proteins were 72.5%,18.6%,0 and 68.8%,15.7%,0,respectively.The expression levels of NF-κB and MMP-9 in CSES were significantly higher than those in CIN and NCE ( P 〈0.05).The expression levels of NF-κB and MMP-9 were correlated to the degree of cell differentiation ,clinical stage and lymph node metastasis of cervical carcinoma ,however,which were not related with patient 's age and histological type.The expression levels of NF-κB were positively correlated with those of MMP-9 ( r =0.872, P 〈0.05).The total effective rate of IC was 81.3%,moreover,the expression levels of NF-κB and MMP-9 after IC were significantly decreased ,as compared with those before IC .Conclusion The overexpression of NF-κB and MMP-9 exists in patients with cervical carcinoma and IC can significantly reduce the expression levels of NF-κB and MMP-9,as a result,which may inhibit the proliferation ,infiltration,invasion and metastasis of tumor cells .
