AMD3100对人肝癌HepG2细胞增殖与AFP表达的抑制作用

摘要 目的:检测AMD3100对人肝癌HepG2细胞增殖及其与甲胎蛋白(alpha fetoprotein,AFP)表达的影响并探讨其意义。方法:显微镜观察不同浓度AMD3100对人肝癌HepG2细胞生长的影响;MTT法检测不同时间AMD3100对人肝癌细胞HepG2增殖的影响,用Western blotting分析不同浓度AMD3100干预前后AFP、CXCR4的表达变化。结果:AMD3100对人肝癌HepG2细胞的增殖有明显的抑制作用,且随着浓度的增加抑制作用越明显;给AMD3100处理后HepG2细胞的CXCR4和AFP表达明显减弱。结论:AMD3100对HepG2细胞的增殖有抑制作用,这种作用很可能与抑制AFP的表达有关。 Objective. To investigate effects of AMD3100 inhibiting the expression of alpha feto- protein and the proliferation of human haptoma HepG2 cells. Methods. The effects of different concentra- tions of AMD3100 on proliferation of human hepatocellular carcinoma cells HepG2 were observed under microscope, and the effects on proliferation of HepG2 in different time points were detected by MTT~ Western blotting method was applied to detect changes of the expression of AFP and CXCR4; Results. It demonstrated that AMD3100 might inhibit proliferation of HepG2 cells, especially at high concentration (〉1.0 μg/mL). It also has function of down-regulation the expression of CXCR4 and AFP in HepG2 cells. Conclusion= AMD3100 inhibited the proliferation of human hepatoma cells, HepG2, and this inhibi tion might be relation with the down-regulation the expression of AFP in human hepatocellular carcinoma cells.
