摘要
通过高效液相色谱法(HPLC)建立一种有效的测定人体血液中哌拉西林浓度的方法,为临床上合理使用哌拉西林提供理论依据。采用WatersC1s柱(250mmx4.6mm,5txm),流动相为乙腈-0.11%四丁基氢氧化铵溶液,卜安息香肟为内标,紫外检测器进行检测。实验结果显示哌拉西林的线性范围为0.5-100μg·mL-1,低、中、高3个浓度日内、日间精密度均小于5%,表明该方法灵敏度高,重现性好,可用于临床中哌拉西林血药浓度的测定以及临床药动学的研究。
The High Performance Liquid Chromatography (HPLC) method was established for the ef- fective determination of piperacillin in human plasma, thus we will provide theory basis for using pip- eracillin in the clinical. A Waters C18 column (250 mmx4.6 mm, 5 ~m) was used with the mobile phase of acetonitrile and 0. 11% tetrabutylammonium hydroxide solution. 1-Benzoin oxime was used as internal standard. Ultraviolet detector was employed. The results showed that the calibration curve of piperacillin was linear in the rang of 0. 5 - 100 Ixg " mL-1. RSDs of the inter-day and intra-day precisions of low, medium and high concentrations were all less than 5%. So the method is sensitive, specific, repeatable and accurate for the determination of piperacillin in human plasma, and is availa- ble for the research about Clinical Pharmacokinetics.
