多叶棘豆总黄酮大孔吸附树脂纯化工艺 被引量:1

Purification technology of total flavonoids from Duoye Jidou (Oxytropis myriophylla) with macroporous resin

摘要 目的研究大孔吸附树脂纯化多叶棘豆总黄酮的工艺条件。方法以总黄酮含量和吸附-解吸率为指标,考察5种不同大孔吸附树脂对总黄酮的吸附能力,确定D4006型树脂为纯化用树脂,对纯化工艺中吸附条件、除杂条件和洗脱条件进行优化。结果最佳工艺条件为上样液浓度0.05 g/mL,最大上样量24倍柱体积(BV),吸附流速3 BV/h,树脂柱径高比1∶9,待吸附结束后,用5%乙醇洗脱9 BV进行除杂,再用50%乙醇以3 BV/h的流速洗脱8 BV,经D4006型大孔吸附树脂纯化后,多叶棘豆总黄酮含量可达54%。结论所建立的工艺分离效果良好,操作简单,重复性好,可用于分离富集多叶棘豆总黄酮。 Objective To study the technology of macroporous resin in the purification of the total flavonoids in Duoye Jidou (Oxytropis myriophylla).Methods Taken the content and adsorptiondesorption rate of the total flavonoids as the indicators,the adsorption ability of 5 kinds of macroporous resins to the total flavonoids were observed,and resin D4006 was determined to be used in purification for optimizing the conditions of adsorption,impurity removing and elution in purification technology.Results The best purification conditions were as follows:the concentration of sample was 0.05 g/mL,maximum sample volume was 24 BV,adsorption flow velocity was 3 BV/h,and ratio of diameter to height was 1∶9.After adsorption,5% ethanol was used in elution (9 BV) for impurity removing and then 50% ethanol was used in elution (8 BV,3 BV/h) again.The content of the total flavonoids reached to 54% after the purification with resin D4006.Conclusion The developed technology has good separation effectiveness and is simple with higher repeatability,which can be used for separating and enriching the total flavonoids in Duoye Jidou.
