大黄灵仙胶囊防治兔胆石病的代谢组学及超微病理研究 被引量:16

Dahuang Lingxian Capsule for prevention and treatment of rabbit cholelithiasis

摘要 目的:用代谢组学及超微病理技术观察大黄灵仙胶囊对兔胆石病的干预作用。方法:将新西兰大白兔60只随机分为正常对照组、药物对照组、模型组和大黄灵仙胶囊治疗组,分别予普通饲料、普通饲料+大黄灵仙胶囊、致石饲料、致石饲料+大黄灵仙胶囊饲养8周。利用电镜观察肝组织超微结构。核磁共振氢谱分析血浆代谢组学特征。结果:与正常对照组比较,模型组低密度脂蛋白、丙氨酸和甘油水平较高;与模型组比较,大黄灵仙胶囊治疗组实验兔血浆低密度脂蛋白、丙氨酸和甘油水平偏低,胆碱、牛磺酸水平则较高。结论:大黄灵仙胶囊使低密度脂蛋白、丙氨酸和甘油的含量降低,并提高胆碱、牛磺酸含量,对影响胆结石发生的血浆代谢产物产生调控作用,并且保护肝细胞功能及结构,降低胆结石的发生率。 Objective:To observe the intervention effect of Dahuang Lingxian Capsule on cholelithiasis by the techniques of metabonomics and ultramicroscopic pathology. Methods: 60 New Zealand rabbits were randomly divided into: normal control group, medication control group, model group, and Dahuang Lingxian Capsule group, fed respectively with common feed, common feed + Dahuang Lingxian Capsule, lithogenic feed, and lithogenic feed + Dahuang Lingxian Capsule for 8 weeks. Electron microscopy was used to observe the ultrastructure of hepatic tissue, and nuclear magnetic resonance hydrogen spectrum (1H NMR) to analyze the metabonomic characteristics plasma. Results: Compared with the normal control group and the medication control group, the model group was relatively high in low density lipoprotein, alanine, and glycerol; and hyperblastosis, cellular edema. Compared with the model group, the Dahuang Lingxian Capsule was relatively low in low density lipoprotein, alanine, and glycerol, and high in choline and taurine. Conclusion: Dahuang Lingxian Capsule decreases low density lipoprotein, alanine, and glycerol, increases choline and taurine, regulated the metabolites of plasma that influenced the incidence of cholelithiasis, protected the function and structure of hepatic cells, and thus reduced the ratio of cholelithiasis.
