木聚糖酶水解竹材废弃物制取低聚木糖工艺条件的优化 被引量:4

Optimization of xylo-oligosaccharides production by xylanase hydrolysis with bamboo wastes in processing

摘要 采用单因素实验优化了竹材废弃物适宜的碱性过氧化氢机械预处理条件,并分别使用单因素和正交实验对竹材废弃物的最佳木聚糖酶水解条件进行了优化。以挤出液的pH及还原糖浓度为指标考察碱性过氧化氢机械预处理效果发现,NaOH用量为1%,H2O2用量为1%,温度为80℃为最佳条件。在单因素实验基础上,采用L9(34)正交实验设计优化了酶水解的工艺条件,最佳工艺条件为:木聚糖酶用量10%、料液比1∶20g/mL、水解时间8h,此条件下酶水解总糖得率为12.11%。 The chemical and mechanical pretreatment conditions of bamboo waste were optimized. The enzyme activity was determined. Bamboo waste xylanase hydrolysis conditions were optimized using single factor and orthogonal experiment. The alkali-hydrogen peroxide mechanical pretreatment conditions were optimized taking pH and reducing sugar as the index. Found that NaOH dosage 1% ,H2O2 dosage 1%,temperature 80℃ were the best conditions. The bamboo waste enzyme hydrolysis process was optimized by L9(3^4) orthogonal experiment on the basis of single factor experiment,the results showed that the optimum conditions were solid-liquid ratio 1:20,enzyme dosage 10%,hydrolysis time 8h. The enzymatic hydrolysis yield of total sugar reached 12.11%.
