非小细胞肺癌患者肿瘤组织与外周血中BRCA1表达的相关性研究

Research on correlation of the BRCA1 expression between tumor tissues and peripheral blood in nonsmall-cell lung cancer patients

摘要 目的检测非小细胞肺癌患者肿瘤组织和外周血中BRCA1 mRNA的表达及相关性,探讨其临床意义。方法应用逆转录-聚合酶链式反应(RT-PCR)法检测35例未经化疗的非小细胞肺癌患者肿瘤组织与外周血中BRCA1 mRNA的表达水平。并以表达水平的中位值作为截断值,将BRCA1的表达程度划分为高表达、低表达。结果肿瘤组织中BRCA1 mRNA的相对表达量为4.00±4.05,中位值为2.3;外周血中相对表达量为2.40±2.33,中位值为1.7。肿瘤组织的相对表达量明显高于外周血,具有统计学意义(P=0.047)。肿瘤组织与外周血中BRCA1 mRNA相对表达量具有相关性(相关系数r=0.245,P=0.041),同时高表达或同时低表达的表达程度相符率达到80%。结论未经化疗的非小细胞肺癌患者外周血在一定程度上可以替代肿瘤组织进行BRCA1表达检测。 Objective To investigate the correlation of breast cancer susceptibility gene 1 ( BRCA1 ) expres- sion in tumor tissue and peripheral blood specimens to explore the feasibility of BRCA1 expression in pe- ripheral blood instead of tumor tissues being molecular marker in non-small-cell lung cancer (NSCLC) chemotherapy. Methods Thirty-five patients with NSCLC underwent surgical resection were recruited. None of the patients had prior chemotherapy. The BRCA1 mRNA expression levels in tumor tissues and peripheral blood were detected by semi-quantitative reverse transcription-PCR (RT-PCR) method. The median level was used as cutoff value to dichotomize patients into high-expression and low-expression BRCA1 sub- groups. Results The average expression levels of BRCA1 mRNA in tumor tissues and peripheral blood were 4.00 ± 4.05 and 2. 40 ± 2.33 ,the median value were 2.3 and 1.7 respectively;the expression of BRCA1 mRNA in tumor tissues was significantly higher than that in peripheral blood ( P = 0. 047 ) ; there was posi- tive correlation between the BRCA1 mRNA expression in tumor tissues and peripheral blood ( r = 0. 245, P =0. 041 );the coincidence rate of the BRCA1 expression grades between tumor tissues and peripheral blood was 80%. Conclusions The expression levels of BRCA1 mRNA in peripheral blood can indirectly re- flect their expression levels in lung tumor tissues.
