饲养的不同环境对局灶性脑梗死大鼠梗死灶周围IGF-1表达的影响

摘要 目的观察饲养的不同环境对局灶性脑梗死大鼠行为学恢复及梗死灶周围胰岛素样生长因子-1(IGF-1)表达的影响,为临床应用不同环境进行脑卒中康复治疗提供基础理论支持和实验依据。方法将实验动物随机分为假手术对照组(15只)和大脑中动脉阻塞(MCAO)(90只)。手术组用电凝法造成右侧大脑中动脉阻塞(MCAO)模型。术后标准笼中群居社交组,5只为一组,共30只;迷宫笼中居学习组,10只一组,共30只;30只居于丰富环境笼。术后剖颅取脑,采用石蜡包埋、切片,免疫组化染色,测梗死灶周围皮质胰岛素样生长因子-1(IGF-1)的表达情况,分别在术后28天、14天、7天、3天、1天随机在各组中选取5只进行,假手术组于术后1天、3天、7天随机选取大鼠5只处死。结果免疫组化结果:IGF-1阳性细胞表达:社交组MCAO术后3 d可见梗死灶周围IGF-1表达增高(P<0.05),7 d时表达略有下降,各手术组与假手术组没有明显差异(P>0.05)。各手术组梗死灶周围皮质IGF-1阳性表达随时间延长逐渐增高,于MCAO术后第14天和28天时各手术组表达均高于假手术组(P<0.05),探索学习组、丰富环境组于MCAO术后IGF-1表达均明显优于社交组在14天和28天(P<0.05)。结论在丰富环境及学习笼中的局灶性脑梗死大鼠,梗死灶周围皮层IGF-1的表达明显上调,可能为功能恢复的原因。 Objective The experiment intended to observe the effects of the different environmental intervention on the behavioral recovery and the expression of insulin - like growth factor - 1, ( IGF - 1 ) around the infarct in rats with unilateral local cerebral infarction, which can offer the basic theoretical support and experimental instruction for stroke rehabilitation. Methods One hundred and five male SD rats, 3 - 4 months, weight 240 - 260 g, were divided into sham group (n = 15 ) and MCAO (middle cerebral artery occlusion) group (n=90). The rats in MCAO group were subjected to electric coagulation of MCA and then were assigned randomly to (1)social communication group ( n = 30, 5 as a group housed in a standard cage) ; (3)learn training group (n = 30, 10 as a group housed in a maze cage) ; (3)rich environment group (n = 30 , housed in a rich environment cage). 5 rats from each operated group at 1 d, 3 d, 7 d, 14 d, and 28 d after MCAO and 5 rats from sham group at 1 d, 3 d, 7 d were randomly assigned to be killed. The expression of IGF - 1 in peri - ischemic cortex were examined from paraffin slice and immuno- histochemistry staining. Results The result of expression of IGF - 1 in the peri - ischemia cortex : the positive cells of IGF - 1 social communicate group is significant higher than sham group on day 3 ( P 〈 0.05 ). The expression of IGF - 1 on MCAO groups slightly decrease on day 7. There are no statistically differences between surgical groups and sham group on day 7 ( P 〉 0.05 ). There are statistically significant differences in MCAO group and sham group on day 14 and 28 ( P 〈0.05). The rich environment and learn traning groups are significantly higher than social communicate group on day 14 and 28 ( P 〈 0. 05 ). Conclusion Learn training and enriched environment after MCAO could up - regulate the expression of IGF - 1 in the peri - ischemic regions. The up - regulation might contribute to the improvement of the recovery of extremity functions resulted from learn training and the enriched environment.
