银杏内酯B对体外培养神经细胞延迟整流钾电流的作用 被引量:1

摘要 目的研究银杏内酯B对传代培养的SH SY5Y细胞延迟整流钾电流(IK)的影响。方法传代培养SH SY5Y细胞,用浓度为0.1、1.0和10.0μmol/L的银杏内酯B灌流细胞,用全细胞膜片钳技术记录SH SY5Y细胞IK。结果浓度为0.1、1.0和10.0μmol/L的银杏苦内酯B在指令电压+60 mV时使SH SY5Y细胞IK峰密度值分别从(2.19±0.45)pA/pF增大至(2.48±0.37)pA/pF(P<0.05,n=5)、(2.97±0.29)pA/pF(P<0.01,n=5)和(3.15±0.48)pA/pF(P<0.01,n=5)。IK分别增大13.24%、35.62%和43.85%,具有浓度依赖性。结论银杏内酯B能增加IK,并且具有浓度依赖性。 Objective To detect the effect of ginkgolide B ( GB ) on delayed rectifier potassium current in subcuhured SH-SY5Y cells. Methods SH-SY5Y cells wele cultured and perfused with GB 0.1,1.0, 10.0 μmol/L. Delayed rectifier potassium current (IK) was recolxted by whole cell patch clamp technique. Results GB at concentrations of 0.1,1.0, and 10.0μmol/L increased IK density value of SH-SY5Y ceils from ( 2.19±0.45 ) pA/pF to ( 2.48±0.37 ) pA/pF ( P 〈 0.05, n =5 ), ( 2.97±0.29 ) pA/pF ( P 〈 0.01, n =5 ), and ( 3.15±0.48 ) pA/pF ( P 〈 0.01, n =5 ), respectively. IA was respectively inereased 13.24%, 35.62% and 43.85% with eoncentration-dependent. Conclusion GB carl significantly increase Ix in a con- cenh-ation-dependent manner.
