肾脾阳虚对实验大鼠血清SOD活力、MDA含量及海马组织形态的影响 被引量:4

摘要 目的采用肌注氢化可的松和灌胃大黄水煎液复合方法复制肾脾阳虚模型大鼠,观察肾脾阳虚对大鼠血清SOD活力、MDA含量及海马组织形态的影响。方法 40只SD大鼠,随机分为正常对照组、模型组、维生素E组和桂附理中丸组。正常对照组大鼠肌肉注射生理盐水,同时灌胃生理盐水,其余组大鼠肌肉注射氢化可的松,同时灌胃大黄水煎液,维生素E组灌胃维生素E混悬液,桂附理中丸组灌胃桂附理中丸混悬液,共30 d。检测血清SOD活力、MDA含量,在透射电镜下观测海马CA1区超微结构的变化。结果肾脾阳虚模型组大鼠血清SOD活力显著低于正常对照组、MDA含量显著高于正常对照组(P<0.01),桂附理中丸组大鼠血清SOD活力明显高于模型组(P<0.05),而MDA水平明显低于模型组(P<0.05)。肾脾阳虚模型大鼠大脑海马CA1区细胞器及神经元出现衰老表现,桂附理中丸组大鼠海马CA1区细胞轮廓清晰,无细胞器及神经元细胞衰老征象,用药效果明显优于维生素E组。结论模型组大鼠出现血清SOD活力降低、MDA含量增多,说明肾脾阳虚可导致氧自由基过量聚积,脂质过氧化终产物增多,进而发生衰老。肾脾阳虚模型大鼠海马细胞衰老的病理改变明显,表明肾脾阳虚可导致大鼠衰老,并与海马神经元细胞受自由基攻击受损密切相关。温补肾脾方药可以提高机体的抗氧化能力,减轻自由基损伤,改善大脑海马组织神经元细胞的病理改变,从而延缓其衰老进程。 Objective Using injected with hydroeortisone and administered with decoction liquid of rhubarb had established deficiency of kidney and spleen - Yang disease models, to observe the serum superoxide dismutase activity and the malonaldehyde content and the hippocampus changes. Methods Totally 40 SD male mice at the age of eight weeks, these mice were randomly divided into normal control group, model group, Vitamin E group, Guifulizhongwan group, ten normal mice in each group, which were sepa- rately given medicine. Excluding normal control group, the mice were injected with hydrocortisone and administered with decoction liquid of Detect the for established deficiency of kidney and spleen - Yang disease models, the procedures lasted for 30 days. De- tect the serum superoxide dismutase activity and the malonaldehyde content; Observation of uhrastructure of hippocam - pus chan- ges. Results Compared with the normal control group, the model group mice of serum SOD activity were significant decreased (P 〈 0.01 ), MDA content was dramatically increased (P 〈 0.01 ) ;compared with the model control group, the Gnifulizhongwan group mice whice of serum SOD activity were obviously increased ( P 〈 0.05 ), MDA content was dramatically decreased ( P 〈 0.05 ). Compared with the normal control group, the model group mice of hippocampal organization organelles were reduced, the neuron atrophy, neurons synaptic gap widening;compared with the model control group, the Vitamin E and Guifulizhongwan group mice of hippocampal organization organelles were increased ,withers as rarely as the neuron atrophy, neurons synaptic gap widening. Conclusion The deficiency of kidney and spleen - Yang disease models mice serum SOD activity was decreased, MDA content was increased ; hippocampal organization organelles has appeared aging. Guifulizhongwan can postpone deficiency of kidney and spleen - Yang causes the aging advancement.
