摘要
以采自峨眉山的桢楠叶和带叶芽的茎为外植体,MS为基础培养基,组合4种不同激素配比的培养基,进行愈伤组织诱导实验,结果表明:(1)4种不同植物激素配比的培养基,都能促使桢楠不同部位的外植体诱导形成愈伤组织,但诱导愈伤组织形成情况具有较明显的差异.(2)以MS为基础培养基,添加2.0mg/L NAA+2.0mg/L 6-BA,是愈伤组织诱导的最佳培养基.(3)以叶为外植体诱导形成愈伤组织的能力强于以带叶芽的茎段,形成的愈伤组织具有颜色偏绿、质地紧密、褐化少等特点,将愈伤组织转接到增殖培养基上,具有增殖能力.
In this study,Phoebe zhennan leaves and stems with buds collected from Emei Mountain were selected as explants,and MS medium combined with four different ratio of hormones were used to conduct callus induction experiments.The results showed that all explants of Phoebe zhennan in this study can be in- duced with callus for the four different ratio of culture medium,but the induction effect of callus has signifi- cant difference among the different parts;the best medium for callus induction is MS medium adding 2.0 mg/L NAA and 2.0mg/L 6-BA;callus induced by leaf as explant is much easier than leaf bud stems,if transferred to proliferation medium, the callus has the ability of proliferation.
