摘要
目的探讨栀子苷对氧嗪酸钾盐致小鼠高尿酸血症的作用及其机制。方法连续7 d灌胃给予氧嗪酸钾盐诱导小鼠高尿酸血症,对比各组血清尿酸水平和24 h尿酸排泄量,检测肝脏黄嘌呤氧化酶(XOD)的活性和肾脏尿酸转运体(mURAT1、mGLUT9、mOAT1)的mRNA和蛋白表达水平。结果栀子苷可显著降低高尿酸血症小鼠血清尿酸水平并升高24 h尿酸排泄量,同时可抑制肝脏XOD活性,调节肾脏尿酸转运体的表达。结论栀子苷对氧嗪酸钾盐致小鼠高尿酸血症具有明显的改善作用,其作用机制可能是通过抑制XOD活性和调节尿酸转运体的表达水平实现。
Objective To investigate the effect of jasminoidin on potassium oxonate-induced hyperuricemia in mice and its mechanism. Methods Hyperuricemia was induced in mice by oral administration of potassium oxonate for 7 consecutive days. Serum uric acid levels and uric acid excretion in 24 h of mice were compared among different groups. Simultaneously, hepatic xanthine oxidase (XOD) activities, as well as mRNA and protein expressions of renal urate transpoters (mURAT1, mGLUT9, mOAT1) were analyzed. Results Jasminoidin significantly lowered the serum uric acid levels and increased the uric acid excretion at 24 h. It remarkably inhibited the hepatic XOD activities and regulated the expressions of renal urate transporters in mice. Conclusion Jasminoidin may obviously improve potassium oxonate-induced hypenaricemia in mice inhibiting hepatic XOD activities and regulating expressions of renal urate transpoters.
