摘要
目的:探讨4种烤瓷金属对人牙龈成纤维细胞(human gingival fibroblast cells,hGFs)增殖及产生炎性因子TNF-α的影响。方法:制备镍铬合金(Ni-Cr)、钴铬合金(Co-Cr)、钛合金(Ti alloy)、纯钛(Ti)浸提液,用ELISA测定hGFs在浸提液中培养1、2、3、6 h后TNF-α的水平;运用MTT比色法比较细胞培养48 h后细胞吸光度值,计算相对增殖率。结果:细胞在浸提液中培养3 h时TNF-α达到高峰,此时Ni-Cr、Co-Cr、Ti alloy中的TNF-α含量明显升高,而Ti与空白组之间差异无显著性;48 h后各组细胞的相对增殖率从小到大依次为Ni-Cr组77.78%、Ti alloy组79.01%、Co-Cr组85.19%和Ti组98.77%,各组的细胞毒性均为0级或1级。结论:Ni-Cr、Co-Cr、Ti alloy可以促进hGFs分泌TNF-α,抑制细胞增殖;而Ti对hGFs增殖及TNF-α分泌无影响。
Objective:To investigate the effects of 4 kinds of dental ceramic alloys on the proliferation and tumor necrosis tactor-α (TNF-α) expression of human gingival fibroblast cells (hGFs).Methods:The leaching liquids of different kinds of dental ceramic alloys were prepared.TNF-α level in the culture medium was measured at different time points by ELISA after cells were cultured with different dental ceramic alloy extractions for 1,2,3 and 6 h respectively; MIT assay was used to evaluate the cytotoxicity of 4 ceramic alloys on hGFs after 48 h cuhure,relative proliferation rate of hGFs was calculated.Results:When hGFs were cultured with leaching liquids of Ni-Cr alloys (Ni-Cr),Co-Cr alloys (Co-Cr) and Titanium alloy(Ti alloy),the expression of TNF-α increased at 1 h after culture,and peaked at the third hour,and then decreased.In Ti extraction TNF-α level was not significantly changed and was not different with that in control medium.After 48 h culture,the relative proliferation rate of hGFs in group of Ni-Cr,Co-Cr,Ti alloy and pure Ti were 77.78%,85.19%,79.01% and 98.77% respectively.Conclusion:Ni-Cr,Co-Cr and Ti alloy can promote TNF-α expression of hGFs,and have an effect on cell proliferation ;Ti has no effect on TNF-α level of hGFs and cell proliferation,indicating that biocompatibility of pure Titanium is better than the others.
