摘要
目的调查中国拉萨藏族无关个体D6S1043、D7S3048、D9s925、D10S2325、D11S2368、D14S608、D15S659、D17S1290、D20S470和GATA198805等10个短串联重复(shorttandemrepeat,STR)基因座的遗传多态性。方法用自行构建的10个常染色体STR基因座荧光复合扩增体系对拉萨藏族208名无关个体的上述10个STR基因座进行遗传多态性分析,计算群体遗传学参数。结果D6S1043、D7S3048、D9S925、D10S2325、D11S2368、D14S608、D15S659、D17S1290、D20S470和GATA198805基因座在拉萨藏族人群分别检出15、10、10、13、10、10、9、11、13和10个等位基因,以及50、41、22、53、34、34、32、31、48和37种基因型。经Bonferroni校正,10个STR基因座基因型频率分布均符合HardyWeinberg平衡,多态性信息含量在0.750~0.860之间,杂合度在0.726~0.865之间,个体识别力在0.919~0.968之间,非父排除概率在0.470~0.725之间,累积非父排除率为0.9998,累积个体识别能力为0.999999999997。结论上述1o个STR基因座在拉萨藏族群体中等位基因频率分布均匀,多态性好,对群体遗传学研究和进行法医学应用具有重要意义,可以作为现有商品化试剂的有益STR补充。
Objective To investigate genetic polymorphisms of 10 short tandem repeats loci (D6S1043, D7S3048, D9S925, D10S2325, Dl1S2368, D14S608, D15S659, D17S1290, D20S470 and GATA198B05) in Tibetans from Lhasa, China. Methods Following extraction, DNA from 208 unrelated Tibetan individuals was amplified with a self-designed multiplex PCR system. The amplified fragments were separated by electrophoresis on an ABI 3130 Genetic Analyzer and analyzed with GeneMapper 3. 2 software. Results The distributions of genotype for the 10 STR loci in the population were in accordance with Hardy-Weinberg equilibrium. The polymorphism information component for the 10 loci was 0. 750- 0. 860, the degree of heterozygosity was 0. 726-0. 865, the discrimination power was 0.9]9-0. 968, and the probability of exclusion was O. 470-0. 725. The combined probability of exclusion and combined discrimination power was 0. 9998 and 0. 999999999997, respectively. Conclusion Above STR loci have high probability of exclusion and discrimination power, which can be used as candidate markers for population genetic research and forensic practice for Tibetans from Lhasa, China.
