摘要
目的观察刺五加皂甙对慢性脑缺血大鼠认知功能的干预效应及其可能机制。方法采用四血管结扎的全脑缺血模型,制模成功后大鼠随机分为脑缺血组、刺五加皂甙组,同时设立假手术组,刺五加皂甙组大鼠分别按剂量25 mg/kg、50 mg/kg、100 mg/kg刺五加皂甙腹腔注射,观察大鼠认知功能变化、免疫组化法分析海马血管内皮生长因子(VEGF)表达、透射电镜下海马超微结构的变化。结果与假手术组比较,脑缺血组大鼠的学习记忆能力明显受损、海马VEGF蛋白表达明显增高(P<0.01);与脑缺血组比较,刺五加皂甙组大鼠认知功能明显改善、海马VEGF表达明显降低(P<0.05)。结论刺五加皂甙可通过抑制VEGF表达显著改善缺血性脑损伤模型大鼠认知功能,呈剂量依赖性。
Objective To explore protective effects of Acanthopanax senticousus saponins on cognitive impairment in rats fol- lowing global cerebral ischemia. Methods The model of global cerebral ischemia was established with the four-vessel occlusion model. 60 male Sprague Dawley rats were randomly divided into sham-operation group, ischemia group and Acanthopanax Senti- cousus saponins group. The rats were injected intraperitoneally with Acanthopanax senticousus saponins at 25 mg/kg,50 mg/kg, and 100 mg/kg once a day. The learning and memorizing ability was measured with the Y maze test. The expression of vascular endothelial growth factor(VEGF) in hippocampus was determined by immunohistochemistry staining. Results The learning and memorizing ability of ischemia groups and the expression of VEGF in rats following global cerebral ischemia were significantly dif- ferent from that of sham-operation groups(P 〈 0.01 ). The significantly improvement of cognitive impairment and the expression of VEGF was observed in rats treated with Acanthopanax senticousus saponins as compared with ischemia groups (P 〈 0.05 ). Con- clusion Acanthopanax senticousus saponins can improve the cognitive impairment in rats following global cerebral isehemia through reducing the expression of VEGF.
