摘要
以苦丁冬青苦丁茶为研究材料,通过柱层析和半制备色谱分离纯化得到6种咖啡酰奎尼酸(CQA)类物质产品,并采用TSKgelODS-80TsQA色谱柱和二极管阵列检测器(DAD),建立一种便捷、灵敏的分析苦丁冬青苦丁茶中cOA类物质的高效液相色谱分析方法。结果表明,通过半制备色谱可分离得到纯度95%以上的6种CQA类单体物质:3-CQA、4-CQA、5-CQA、3,4-diCQA、3,5-diCQA和4,5-diCQA。色谱条件:TSKgelODS-80TsQA色谱柱,流动相为水.甲醇-5%。甲酸体系,梯度洗脱,流速0.5mL/min,柱温40℃,检测波长326nm。该方法在一定范围内有良好线性,相关系数为0.9992-0.9999,平均加标回收率为96.34%,相对标准偏差均小于3%。
Six caffeoylquinic acid (CQA) derivatives were separated by column chromatography and semi-preparative high performance liquid chromatography (HPLC) from the crude extract of llex kudingcha C.J. Tseng. An HPLC method with DAD and TSKgel ODS-80 TsQA column was developed for the simultaneous determination of CQAs in Kuding- cha made from llex kudingcha C.J. Tseng. The results demonstrated that six CQA derivatives with 95% purity, 3-CQA, 4-CQA, 5-CQA, 3,4-diCQA, 3,5-diCQA and 4,5-diCQA, could be obtained through the purification of Ilex kudingcha C.J. Tseng by semi-preparative high performance liquid chromatography (HPLC). The chromatographic conditions were determined as follows: oven temperature set at 40 ℃, gradient elution with methanol/formic acid (5%0) with a flow rate 0.5 mL/min, and detection at a wavelength of 326 nm. As results, good linearity was observed with correlation coefficients of 0.9992-0.9999, the average recovery of the CQA derivatives from spiked samples was 96.34%, with relative standard deviations (RSD) less than 3% (n = 3).
