摘要
目的 :了解小儿急性白血病的免疫机理 ,探讨细胞因子和Fas系统在小儿白血病中的作用 ,以便为临床免疫提供可靠的理论依据。方法 :应用双抗体夹心酶联免疫吸附法检测了 39例急性白血病患儿化疗前后血清IFN γ及sFas水平 ,与对照组比较并对IFN γ与sFas进行了相关分析。结果 :①化疗前白血病患儿血清IFN γ水平明显降低 (P <0 .0 1) ;sFas水平明显升高 (P <0 .0 1) ,且女性较男性血清sFas升高更明显。②化疗后达完全缓解时 ,血清IFN γ水平上升 ,而sFas水平下降 ,与正常对照无明显差异 (P >0 .0 5 )。③IFN γ及sFas之间未证实有明显相关性 (r = 0 0 79,P >0 .0 5 )。结论 :急性白血病免疫机理与细胞因子功能失调及凋亡功能障碍有密切关系 ,这些生物因子的异常与白血病的发生、发展有一定关系。
Objective:To investigate the immunopathologic mechanisms and the roles of interferon γ(IFN γ) and a soluble form of Fas(sFas) in children with acute leukemia.Methods:Serum levels of IFN γ and sFas were determined before and after the treatment respectively by enzyme linked immunosorbent assays(ELISA) and analysed by linear correlation in 39 children with acute leukemia,and compared with normal control group.Results:①the level of sFas was significantly higher( P <0.01),while the level of IFN γ was significantly lower( P <0.01) in most acute leukemia(AL) patients at the time of the diagnosis or relapse than the control subjects.Serum level of sFas was significantly decreased and the level of IFN γ was significantly increased in the children of complete remission(CR).②There was no any correlation between IFN γ and sFas( r = 0.079, P >0.05).Conclusion:the immunopathologic mechanisms of children acute leukemia were associated with functional disorders of cytokine and mechanisms of children acute leukemia were associated with functional disorders of cytokine as well as functional disorders of cellular apoptosis. \ \
