摘要
草莓茎尖脱毒培养中茎尖分生组织经历了一个生长发育的动态过程 ,茎尖分生组织在培养 1周左右膨大 ,转绿 ;2~ 4周大量发生愈伤组织且芽突起陆续分化成小苗 ;丛生芽的抽生主要集中在接种后的 5~ 7周。不同草莓品种 ,其茎尖分生组织诱导生长情况不同。在相同条件下 ,鬼怒甘品种的茎尖生长速度快于丰香品种 ,长势强于丰香品种 ,而其丛生芽发生率低于后者。丰香草莓茎尖用MS作基本培养基 ,添加 0 1mg/L的NAA ,在BA浓度为 0 5~ 1 0mg/L范围内均可有效诱导愈伤组织发生和促进芽的分化。丰香草莓茎尖二次脱毒培养的出愈率 (98 0 4% )、直接成芽率 (93 47% )均高于相同条件的茎尖一次脱毒培养 (出愈率为 74 44 % ,直接成芽率为 71 11% )
Shoot apical meristems would undergo a series of changes as the length of culture period lasted. Shoot apical meristems, fllowing culture 3~7 days after inoculation, became big and turned green. They then formed prolific calluses and buds after 2~4 weeks culture. After 5~7 weeks culture multiple buds were produced. Shoot tip growth was affected by variety. In comparison with Fengxiang, shoot tip explants of Gui Nugan grew much faster, while the rate of multiple bud differentiation was lower under same culture conditions. BA concentrations at 0 5~1 0mg/L were effective in inducing the growth of callus and multiple buds. Both the rate of callus formation (98 04%) and that of bud differentiation (93 47%) of the second shoot tip culture were much higher than those of the first shoot tip culture (74 44% and 71 11%, respectively).
