摘要
以鲤鱼为试验材料,研究氟化钠对其脑组织中白细胞介素-1β(IL-1β)蛋白表达的影响。结果显示,在不同浓度(40、80、120 mg/L)氟化钠暴露30、60、90 d后,与对照(氟化钠0 mg/L)相比,鲤鱼血液中、脑组织中的氟含量都有增加,且呈一定的时间-剂量效应关系。在氟暴露90 d时,采用免疫印迹方法检测各组脑组织中IL-1β的表达情况,并测定脑组织中的T-AOC含量,结果显示,与对照相比,不同剂量氟暴露组鲤鱼脑组织中的IL-1β蛋白表达有增加趋势,随着氟剂量的增大T-AOC含量呈下降趋势。CORREL相关分析结果显示,血液中、脑组织氟含量与IL-1β蛋白的表达均呈正相关(r=0.975,r=0.931),与T-AOC含量呈负相关(r=-0.972)。表明鲤鱼血液中氟含量、脑组织中氟含量的上升,可引起脑组织中T-AOC含量的下降和IL-1β表达的上调,进而可能损伤神经系统。
Effects of sodium fluoride on expression of interleukin 1 beta （IL-1β） protein in brain tissue in carp were studied. The results showed that after exposure to different concentrations of sodium fluoride （0, 40, 80 and 120 mg/L） for 30, 60, 90 d, fluoride levels in blood and brain increased with the exposure dose and time increasing, showing time and dose-response relationship. After 90 d of exposure, the expression of IL-1β protein and the T-AOC content in brain tissue were determined, and the results indicated that IL-1β protein expression increased while the T-AOC content decreased in the brain tissue with the increase of fluoride dose. Pearson correlation analysis showed that significantly positive correlation was observed between the expression of IL-1β protein and the fluoride content in blood and brain （r=0.975, r=0.931） while negative correlation between the expression of IL-1β protein and T-AOC content （r=-0.972）, between the fluoride content in brain and T-AOC content （r=-0.924）. All above indicated that the increase of fluoride content in blood and brain damaged the nervous system by increasing the expression of IL-I~ protein and decreasing the T-AOC content in brain tissue.
