摘要
为了将GHSR基因应用于山羊生长性状的标记辅助选择中,利用PCR-SSCP技术分析了黔北麻羊生长激素促分泌素受体GHSR基因外显子1、2、3’UTR区及5’UTR区的多态性及其与黔北麻羊生长性状的关联。结果表明,在GHSR基因外显子2和3’UTR区各检测到2个SNP位点G996A和T1424C,其中G996A位点为错义突变,氨基酸由甘氨酸变为丝基酸,表现为3种基因型,分别命名为GG、GA和AA;外显子1和5’UTR区未发现多态位点。进一步将G996A位点与生长性状进行关联性分析,发现GG和GA基因型个体的体重显著高于AA基因型个体,胸深显著低于AA基因型个体。初步推断GHSR基因G996A位点是非常有价值的辅助选择标记,可为黔北麻羊生长性状方向选择提供理论依据。
In order to increase the possibility of application of growth hormone secretagogue receptor (GHSR) gene in MAS (Marker-assisted selection), PCR-SSCP technology was applied to detect polymorphisms of Exonl, 2, 3'UTR and 5' UTR of GHSR gene in Qianbei Ma goat, and their associations with body weight and body size were analyzed. The result showed that G996A and T1424C were detected in Exon2 and 3 'UTR, whereas, there was no SNPs in Exonl and 5 'UTR of GHSR gene in Qianbei Ma goat. The SNP (G/A) was a missense mutation, which made Gly to Ser and was defined as three genotypes: GG, GA and AA. The study on growth traits and GHSR gene polymorphism revealed that the body weight of individuals with genotype GG and GA were significantly higher than that of genotype AA, smaller than those for chest width. It was concluded that G996A site in GHSR gene was a significant molecular genetic marker of MAS, providing a theoretical basis for selecting growth traits of Qianbei Ma goat.
