摘要
研究了胆固醇氧化酶酶学特性,并对其产生菌COX4-8进行了分类鉴定.经过细菌形态、生理生化以及16S rDNA分子鉴定,确定其为红球菌属(Rhodococcuse sp.).通过对菌株COD4-8来源的胆固醇氧化酶的酶学性质进行研究,结果表明该胆固醇氧化酶的最适pH值为7.0,最适温度为40℃.该酶催化胆固醇的米氏常数K m为3.5×10-6mol/L.Hg2+抑制酶活的能力最强,Mn2+对酶活的影响最小;而SDS、Tween-80和TritonX-100对酶活力的影响也较小.
A bacteria strain named COX4-8,with high level of extracellular cholesterol oxidase (COD) production,was obtained.According to the strain morphology,physiological and hiochemistrical properties,and 16S rDNA sequence of COX4-8,it was identified as Rhodococcuse sp..The properties of cholesterol oxidase from Rhodococcuse sp.were investigated.The optimum activity was obtained at pH7.0 and 40 ℃.The Km value for oxidation of cholesterol by this enzyme was 3.5 x 10-6 mol/L.The studies of the effects of some metal ion and organic compounds on the enzyme activity indicated its potential as a clinical diagnostic reagent.
