细胞外信号调控激酶在尿道下裂发病机制中的作用

Role of extracellular signal-regulated kinase in the pathogenesis of hypospadias

摘要 目的研究细胞外信号调控激酶(extracellularsignalregulatedkinase,ERK)1/2在正常和尿道下裂患儿包皮组织中的表达,探讨其在尿道下裂发病机制中可能发挥的作用。方法收集拟行尿道下裂尿道修复术患儿术中切除的包皮组织(30例)和门诊包皮环切术中切除的儿童包皮(30例),采用免疫组织化学和实时定量聚合酶链反应(qPCR)方法检测两组包皮组织中ERK1和ERK2mRNA和蛋白质的表达变化。结果尿道下裂患儿包皮组织中ERK2mRNA表达较正常水平有显著升高,差异有统计学意义(P〈0.05)。总ERK1/2和磷酸化ERK1/2蛋白主要表达于包皮皮下问充质细胞层,其中磷酸化ERK1/2蛋白在尿道下裂患儿包皮组织的表达较正常对照有明显增强趋势。结论ERK1/2的表达异常与尿道下裂发生相关,其介导的空间调控异常可能是导致尿道下裂发生的重要机制之一。 Objective To investigate the expression of extracellular signal-regulated kinase (ERK) 1/2 in foreskin tissue from boys with hypospadias in order to determine its role in the patho genesis of hypospadias. Methods Penile skin tissues obtained at hypospadias repair (30 cases) ztnd e- lective circumcision (3tl cases) were studied. Real-time quantitative polymerase chain reaction (qPCR) and immunoehemistry were employed to determine the mRNA expression and protein phosphorylation level of ERKI and ERK2. Results Compared to normal control, the expression of ERK2 mRNA in hypospadias patients foreskin tissue increased significantly(P〈0.05). ERK1 and 2 total and phospho- rylated protein were mainly expressed at the preputial subcutaneous mesenchymal cell layers. The phosphorylation protein levels of ERK1/2 in foreskin from children with hypospadias showed an in- creasing trend. Conclusions Our results indicate that the aberrated ERK1/2 may be associated with the formation of hypospadias through its spatial regulation.
