摘要
目的 :探讨络通对糖尿病大鼠视网膜的作用及其可能机理。方法 :40只SD大鼠随机分成对照组、链脲佐菌素糖尿病组与络通预防组 ,络通粉末 4g/(kg·d)混入普通饲料给药。1 80d后处死动物 ,取血检测血浆一氧化氮、丙二醛、全血谷胱甘肽过氧化物酶、组织型纤溶酶原激活物、纤溶酶原激活剂抑制物等生化指标及红细胞压积、全血粘度、纤维蛋白原等血液流变学指标 ,并在光镜下进行视网膜消化铺片血管形态学观察及周细胞记数。结果 :糖尿病大鼠存在明显的高凝、高粘现象 ,并出现了周细胞减少、微血管瘤、无细胞毛细血管及影周细胞等特征性糖尿病视网膜病变 ;络通干预后 ,高凝、高粘现象及视网膜病变明显改善。结论 :络通对早期糖尿病大鼠视网膜病变具有良好的保护作用 ,这可能与其能减轻氧自由基损伤 。
Objective:To evaluate the protective effect and mechanism of action of luotong on streptozotocin induced diabetic retinopathy. Methods:Forty male Sprague Dawley rats were randomly divided into normal control rats,streptozotocin diabetic rats and diabetic rats treated with luotong .The powder of luotong 4 g/kg·d were mixed with food.All rats were killed after 6 months.Hemodynamics,nitrogen monoxide(NO),malondialdehyde(MDA),glutathione peroxidase(GSH PX),t PA and PAI were observed.Retinal digest preparations were observed with light microscope,and the number of pericyte was counted.Results:After six months,the STZ diabetic rats retinal digest preparations showed characteristic retinal changes including decrease of pericytes,capillary microaneurysms,acellular capillary and ghost pericyte.Retinal changes of luotong preventive group were less severe than that of model group,and so were various indexs.Conclusion:Early treatment of luotong may be helpful in delaying or possibily preventing against the diabetic retinopathy,and this effect may be partly achieved by decreasing the lesions of oxygen free radical,correcting the disturbance of hemorrheology and plasma t PA,PAI activities.
