摘要
目的制备用于研究风疹病毒(RV)生物学特性及血清学应用的特异性McAb。方法RV抗原免疫BALB/c小鼠,无菌取其脾细胞与小鼠骨髓瘤细胞SP_2/O融合建立杂交瘤细胞,并对杂交瘤细胞产生抗体特性及相应McAb进行了分析。结果成功建立了三株稳定分泌McAb的杂交瘤细胞5D_5、5G_5、2F_7,染色体数目均大于98条;相应McAbs均属IgG_1,与纯化RV抗原有强阳性反应。结论该法可产出高特异性、高亲和力RV特异性McAb。
Objective The study is for preparing the specific monoclonal antibody of rubella virus(RV) which could be applied for researching the biological character of RV and diagnesing human infection by it. Methods BALB/c mice were immunized by RV antigen and their spleen cells were estheted to fuse with the mice myeloma cells SP_2/O to es tablish hybridoma cell strain.The hybridoma cells and homologous monoclonal antibody were analyzed by experiments. Re sults Establishing three-strain hybridoma cells, 5D_5、5G_5 and 2F_7 sueccessfully, which secret monoclonal antibody stably.The number of chromatosome is all over 98 streaks. The homologous monoclonal antiodes all belong to IgG_1 .They react with strong masculine on RV pare antigen. Conclusion The methods establied by the experiments can produce meno clonal antibody with hyper-affinity and specificity of RV.
