应用XMP-2型松材线虫快速分离器分离松木材中线虫的试验 被引量:2

Isolating Nematodes from Pine Board Using XMP-2 Type of Quick Isolation Instrument for Bursaphelenchus xylophilus

摘要 应用XMP-2型松材线虫快速分离器分离松木板中的线虫,样品准备好后,在1 min左右就从松木板中分离出了线虫,与传统的贝尔曼漏斗法一般需24 h相比,大大地缩短了线虫的分离时间.应用快速分离器在23~77 s内松材线虫的检出率达100%,每块样品平均检出线虫47只,初次分离得到的线虫数足够用于检疫检验;初次分离时浸湿的松木板放置10 h再用该仪器分离,可得到更多的线虫.由于松材线虫是林业高度危险的传染性病害,病原线虫的快速检出对准确判断病情及病情的快速、有效控制具有重要意义,因此XMP-2型松材线虫快速分离器可广泛应用于松木材的检验检疫,以提高工作效率. Bursaphelenchus xylophilus is a highly-dangerous infectious disease of pine forest,so rapid isolation of pathogen nematodes is very important to accurately judging and efficiently controlling of the disease.In this communication report,results of isolating nematodes from pine board using XMP-2 type of quick isolation instrument for B.xylophilus invented by Xiamen University,were reported.The instrument was designed and manufactured based on principle of pressure difference,and nematodes could be rapidly carried out from vasculature by stream washing through pine board,by the suction force created by negative pressure cavity.The results were compared with the classical method of isolating nematodes,Baermann funnel method (gravity principle),which has been used for more than a hundred years.Nematodes in pinewood sample could be isolated in one minute using quick isolation instrument while Baermann funnel method required 24 hours,meant this instrument could significantly save isolating time.During 23 to 77 seconds,the nematodes detection rate from pinewood was 100%.The instrument could isolate 47 nematodes averagely from each pinewood sample.Those nematodes were enough for quarantine and examining.More nematodes could be detected from damp pinewood sample which have been wetted for 10 hours after initial isolating.Therefore,XMP-2 type of quick isolation instrument for B.xylophilus could be widely used for pinewood inspection and quarantine,which could improve work efficiency significantly.
