摘要
为了明确乳牛隐性乳房炎诊断液的诊断机理,试验分别以兔血和鸡血作为体细胞样本,配制不同含量的体细胞悬液,使其中的体细胞数与乳牛隐性乳房炎阴性(-)、弱阳性(+)、阳性(++),和强阳性(+++)乳汁中所含的体细胞数一致,并进行测试对比及冻融试验。结果表明:以阴离子表面活性剂为主要成分的隐性乳房炎诊断液具备必要的诊断特异性,其诊断机理主要是阴离子表面活性剂破坏乳汁体细胞,释放细胞核中的DNA,遇水形成高黏度DNA水化物。说明以兔血制备的体细胞悬液,只有白细胞能与隐性乳房炎诊断液反应,与乳汁体细胞反应相同,适合乳汁体细胞研究;以鸡血制备的体细胞悬液,红细胞也能与隐性乳房炎诊断液反应,与乳汁体细胞反应不相同,不适合乳汁体细胞研究。
To clear up the diagnosis mechanism of diagnosis liquid for subclinical mastitis in dairy cows, rabbit and chicken blood was used as somatic cell samples to prepare different contents of somatic cell suspensions, in which their somatic cell counts were consistent with those of milk produced by cows with subclinical mastitis showing negative ( - ), weakly positive ( + ), positive ( + + ), and strongly positive ( + + + ). And then the experiments of test comparison and freeze - thaw were carried out. The results showed that the diagnosis liquid for subclini- cal mastiffs which consisted anionic surfaetant as main ingredient had the necessary specificity for the diagnosis, its diagnosis mechanism was mainly that anionic surfaetant damaged milk somatic cells to release the DNA in the nucleus, and then the DNA contacted with water to form DNA hydrate with high viscosity. The results indicate that for somatic cell suspension of rabbit blood, only leukocytes can react with the diagno- sis liquid, and the result is same as that of milk somatic cell response, thus, the somatic cell suspension of rabbit blood is suitable for the study of milk somatic cell. For the somatic cell suspension of chicken blood, red blood cells can also react with the diagnosis liquid, but the result is different from that of milk somatic cell response, thus, the somatic cell suspension of chicken blood is not be suitable for the study of milk so- matic cell.
