消肿止痛合剂对大鼠骨折愈合中VEGF表达的影响 被引量:13

摘要 目的:探讨大鼠骨折修复过程中骨痂组织中血管内皮细胞生长因子(VEGF)表达的变化及消肿止痛合剂对其的影响。方法:取5-6周龄SD大鼠30只,制作股骨中段闭合骨折模型,随机分为实验组和对照组各15只,实验组每12小时予消肿止痛合剂16mL/(kg·d),对照组不予药物治疗。分别在骨折后第2、4、7、10、21天每组各处死3只大鼠,取骨折周围软组织及骨痂,观察骨折部位骨痂形成情况,通过组织切片免疫组织化学染色,观察骨折修复中不同时期软骨细胞变化及骨痂中VEGF的表达。结果:VEGF的表达在2组动物骨折后第4天开始出现差异,第10天差异最大,第21天差异缩小,实验组中VEGF的表达信号强于对照组。结论:骨折愈合过程中软骨及骨性骨痂中均有VEGF表达,表明VEGF参与骨折愈合,消肿止痛合剂能提高VEGF的表达,利于骨折修复。 Objective: To explore the changes of vascular endothelial growth factor (VEGF) expressions in the callus during the healing course of bone fracture and the effects ofXiaoZhong ZhiTong mixture on VEGF expressions of rats. Method: All 30 SD rats were chosen to establish the models, they were randomized into the experiment group and the control group equally, the experiment group took XiooZhong ZhiTong mixture every 12 hours in the dose of 16 mL/( kg. d) and the control group were given with nothing. The rats were killed at the 2nd, 4th, 7th, 10th and 21 st days respectively after fractured, soft tissue and callus around the location of bone fracture were taken to observe the formation of the callus. Changes of cartilage cells in different stages and VEGF expression in the callus were observed in the healing course of bone fracture by immunohistochemistry. Result: The difference appeared between both groups in VEGF expression at the 4th day, the difference was the most significant at the 10th day, it reduced at the 21st day. VEGF expressions in the experiment group was more than that of the control group. Conclusion: VEGF expressions occur in the cartilage and the callus in the healing course of bone fracture, which demonstrates the participation of VEGF in the healing course of bone fracture, XiaoZhong ZhiTong mixture could improve VEGF expression and is helpful to the healing of bone fracture.
