摄入过量氟实验大鼠肾脏形态损害与体内氟含量及血清MDA的关系

摘要 目的 探讨氟中毒肾脏病理形态损害发生的机理。方法  70只 4~ 5周龄 ,体重 50~90 g的Wistar大鼠随机分为对照组、低、中、高剂量氟组 ,观察 3月与 5月。光镜观察各实验组大鼠肾脏病理形态学变化 ,血清MDA含量用硫代巴比妥酸法测定 ,血清氟用氟离子选择电极法测定。结果 摄入过量氟引起肾脏的病理变化为皮质及髓质呈灶状散在分布的肾小管上皮细胞坏死、扩张 ,间质纤维母细胞及胶原纤维灶状增生。随摄入过量氟的浓度增高 ,肾损害率由58 33%增高到 90 0 0 %。与肾损害率密切相关的因素是肾氟含量。 Objective To understand the pathogenic mechanism of the pathologic changes of kidneys in fluorosis.Methods Seventy Wister rats,4~5 week old,body weight 50~90g,were randomly divided into the control,the low dose excess fluoride group,the middle dose excess fluoride group and the high dose excess fluoride group. The experimental period was 3 months and 5 months.The pathologic changes of kidneys were observed by microscope,MDA in serum were determined by sulfobabitoric acid and fluoride contents in serum and bone were assayed by ionization,respectively.Results The pathogenic changes of kidneys in rats treated with excess fluoride were necrosis and dilation in epithelium of renal tubules,interstitial fibroblast and collagen fibril,which scattered in cortex and medulla of kidneys.The damaged rates of kidneys in rats were from 58.33% to 90.91% with the ingested doses of excess fluoride.The factors related with the damaged rates of kidneys were the fluoride content in kidneys.Conclusion Excess fluoride contents that silted up in kidneys is a factor that directly caused the damages of renal tubules in cortex and medulla of kidneys.
