摘要
目的:探讨MMP-9和TIMP-1mRNA在食管鳞状细胞癌和对应癌旁组织中的表达差异及其相关性。方法:选取新鲜手术切除的食管癌标本98例,每例分别取癌组织和对应的癌旁组织各1份,采用RT-PCR法检测MMP-9和TIMP-1mRNA在食管鳞状细胞癌组织和对应癌旁组织中的表达,分析食管癌组织中MMP-9和TIMP-1mRNA表达与临床病理特征的关系;采用Pearson相关分析法分析MMP-9与TIMP-1mRNA在食管鳞状细胞癌组织中表达率的相关性。结果:食管癌组织中MMP-9mRNA阳性表达率高于癌旁组织,阳性表达率比较差异有统计学意义(χ2=22.873,P=0.000);食管癌组织中TIMP-1mRNA阳性表达率高于癌旁组织,阳性表达率比较差异有统计学意义(χ2=7.864,P=0.005)。MMP-9mRNA表达率与食管鳞状细胞癌浸润深度有关联(χ2=5.022,P=0.025),与食管鳞状细胞癌淋巴结转移无关联(χ2=0.617,P=0.432)。TIMP-1mRNA的表达与食管鳞状细胞癌浸润深度有关联(χ2=4.982,P=0.026),与食管鳞状细胞癌淋巴结转移无关联(χ2=0.527,P=0.468)。MMP-9mRNA与TIMP-1mRNA在食管鳞状细胞癌组织中的表达率呈正相关关系(r=2.237,P=0.028)。结论:MMP-9和TIMP-1mRNA表达可能参与了食管鳞状细胞癌的发生发展,但在食管鳞状细胞癌淋巴结转移方面可能不发挥关键作用。
Objective To explore the differences of the expressions of MMP-9 and TIMP-1 mRNA in esophageal squamous cell carcinoma tissue and the matched adjacent normal tissue and their relationships. Methods 98 spicemens of surgical removed esophageal carcinoma tissue were chosen for detection, the carcinoma tissue and the adjacent normal tissue were cut and stored simultaneously from every spicemen, respectively. RT- PCR method was used to detect the expressions of MMP-9 and TIMP-1 mRNA in esophageal squamous cell carcinoma tissue and the adjacent normal tissue; the relationships between the expressions of MMP-9 and TIMP-1 mRNA in esophageal squamous cell carcinoma tissue and clinicopathological characteristics were analyzed. The relationship between the expressions of MMP-9 and TIMP-1 mRNA in carcinoma tissue was analyzed by Pearson related analysis method. Results The positive expression rate of MMP-9 mRNA in carcinoma tissue was higher than that in adjacent normal tissue (X2 =22. 873, P=0. 000), and the positive expression rate of TIMP-1 mRNA in carcinoma tissue was higher than that in adjacent normal tissue (X^2= 7. 864, P= 0. 005). There was correlation between the positive expression of MMP-9 mRNA and the depth of tumor infiltration (X2 =5. 022, P=0. 025), but there was no correlation between the positive expression of MMP 9 mRNA and lymph node metastasis (X2 =0. 617, P=0. 432). There was correlation between the positive expression of TIMP-1 mRNA and the depth of tumor infiltration (X2= 4. 982, P=0. 026), but there was no correlation between the positive expression of TIMP-1 mRNA and lymph node metastasis (X^2= 0. 527, P = 0. 468). The positive expression of MMP-9 mRNA in esophageal carcinoma tissue had positive correlation with the expression of TIMP-1 mRNA (r= 2. 237, P = 0. 028). Oonclusion The expressions of MMP 9 and TIMP-1 mRNA may play a very important role in the development of esophageal squamous cell carcinoma, however both of them have no affection on lymph node metastasis of esophageal squamous cell carcinoma.
