摘要
目的:建立具有稳定浓度梯度的运动神经元体外诱向生长模型。方法:应用神经细胞诱向小室建立生长模型,ELISA法检测模型中不同分区琼脂糖内神经因子浓度;细胞培养法鉴定小室中神经细胞活性以及运动神经元诱向因子对其诱向生长作用。结果:通过诱向小室内运动神经元诱向因子浓度梯度形成,成功建立运动神经元体外诱向生长模型;细胞培养显示诱向小室琼脂糖无明显细胞毒性,具有浓度梯度的运动神经元诱向因子对其有明显诱向生长作用。结论:该生长模型具有良好的可重复性和应用稳定性,可用于体外不同因子对运动神经元诱向生长作用的研究。
Objective To establish a motor neuron-induced regeneration model with the stable concentration gradient of neurotrophic factors in vitro. Methods Establishment of the motor neurons-induced regeneration model was based on the protocol of Cao et al by using nerve cells -induced chamber. In order to confirm the formation of a concentration gradient in the chamber, the concentration of neurotrophic factors were determined in the different partition of agarose by ELISA. The activity of the motor neurons in the chamber and the efficacy of the induced regeneration by cell culture were assessed. Results The concentration gradient and the effect of motor neuron- induced regeneration were observeded in the chamber. The agarose in the chamber had no cytotoxicity, this nerve cell induced regeneration model was successfully established and the model is feasible for the motor neurons. Conclusion A motor neuron-induced growth model was successfully established, which can be used to study the effects of different neurotropbic factors in motor neurons-induced regeneration.
