电针“丰隆”穴对高脂血症大鼠血脂及巨噬细胞肿瘤坏死因子-α、白介素-6的影响 被引量:7

Effect of Electroacupuncture of“Fenglong”(ST 40)on Levels of Blood Lipid and Macrophage TNF-αand IL-6 in Hyperlipidemic Rats

摘要 目的:观察电针"丰隆"穴对高脂血症大鼠炎性反应因子的影响,探讨电针在治疗高脂血症中的作用机制。方法:成年雄性SD大鼠50只,随机分为正常组、模型组、饮食控制组、电针组、饮食控制+电针组,每组10只。用高脂饲料喂养法造模。造模成功后,饮食控制组改喂普通饲料;电针组取双侧"丰隆"穴治疗,每次30min,1次/d,连续治疗30d。治疗结束后,测定各组血脂含量,包括总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C);分离大鼠腹腔巨噬细胞(Mφ),应用流式细胞仪检测肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)的百分含量。结果:模型组大鼠血浆TC、LDL-C较正常组明显上升(P<0.01),HDL-C明显下降(P<0.01),而TG变化不明显(P>0.05);饮食控制组、电针组大鼠血浆TC、LDL-C与模型组比较明显下降(P<0.01),饮食控制+电针组大鼠血浆中TC、LDL-C含量较模型组、饮食控制组明显降低(P<0.01)。模型组、饮食控制组大鼠腹腔Mφ百分含量较模型组明显增加(P<0.01);电针组、饮食控制+电针组大鼠腹腔Mφ百分含量比模型组明显降低(P<0.05,P<0.01);饮食控制+电针组大鼠腹腔Mφ百分含量较饮食控制组显著降低(P<0.05)。模型组大鼠腹腔Mφ内TNF-α、IL-6百分含量与正常组比较均有所增加(P<0.01);饮食控制组、电针组、饮食控制+电针组大鼠腹腔Mφ中TNF-α、IL-6的百分含量均比模型组显著降低(P<0.05,P<0.01);与饮食控制组比较,饮食控制+电针组大鼠腹腔Mφ中TNF-α、IL-6的百分含量显著降低(P<0.01,P<0.05)。结论:电针"丰隆"穴能够明显下调高脂血症大鼠血浆TC、LDL-C水平,该作用与下调腹腔Mφ内炎性反应因子TNF-α、IL-6的表达水平有关。 [ABSTRACT] Objective To observe the effect of electroacupuncture (EA) stimulation of "Fenglong" (ST 40) on blood lipid contents and inflammatory factor levels in hyperlipemia rats so as ta elucidate.its mechanism underlying improvement of hy- perlipemia. Methods Fifty male SD rats were randomized into 5 groups= normal, model, diet -control, EA intervention (EA), diet-control+ EA groups, with 10 rats in each group. Hyperlipemia model was established by feeding the animals with high-fat diet for 30 days. After modeling, rats in the diet-control group were fed with routine fodder. EA was applied to bilateral "Feng- long" (ST 40) for 30 min, once daily for 30 days. Following intraperitoneal injection of 1640 culture fluid, the peritoneal fluid was collected and centrifuged for extracting macrophages. Flow cytometry (FOM) was employed to determine the levels of tumor nec- rosis factor alpha (TNF-a) and interleukin-6 (IL-6) after adding fluoresce-labeled antibodies. Results The contents of serum TC and LDL-C were remarkably higher and HDL-C level was significantly lower in the model group than in the normal group (P〈0.01). After EA intervention, serum TO and LDL-C showed an apparent decrease (P〈0.01). Compared with the normalgroup, percentages of CD11 b, TNF-a and IL-6 were significantly increased in the model group (P^0.01), while in comparison with the model group, percentages of CD11 b in both EA and diet-control + EA groups, TNF-a and I L-6 percentages of macropha- ges in the diet-control, EA and diet-control+ EA groups were notably decreased (P^0.05, P^0.01). The effects of the diet- control+ EA group were considerably superior to those of the diet-control group in lowering CD I 1 b, TNF-a and IL-6 percentages (P〈0.05, P〈0.01). No significant differences were found between the diet-control and EA groups in the aforementioned inde- xes (P〈0.05). CD11 b level indicates changes of macrophage level due to its specific marker character. Conclusion EA stimulation of "Fenglong" (ST 40) is effective in lowering serum TO, LDL-C, and macrophage TNF-a and IL-6 levels in hyper- lipemia rats.
