下颌骨骨痂组织中TGF-β_1及BMP-2的表达及意义 被引量:3

Expression and significance of TGF-β_1 and BMP-2 in mandibular callus

摘要 目的:观察转化生长因子β1(TGF-β1)、骨形成蛋白-2(BMP-2)在人下颌骨骨折骨痂组织中的表达及量的变化。方法:在下颌骨骨折的手术患者中,收集术中去除的骨断端间骨痂组织30例,以2例方颌畸形患者术中切取的下颌角骨块作正常对照;利用SABC免疫组织化学方法检测骨折后不同时期骨痂组织及正常下颌角骨组织中TGF-β1、BMP-2的表达。结果:TGF-β1、BMP-2在正常骨组织中没有表达;在不同时期骨痂组织中均有表达,TGF-β1在骨折后第1周到第3周表达量缓慢增加,至第3周时达到高峰,随着时间推移,表达量逐渐下降;BMP-2在骨折后前2周表达呈增加趋势,第2周达到高峰,以后随时间推移,表达量缓慢下降。结论:①BMP-2可能是启动骨折修复的因子之一;②TGF-β1可能是骨折修复的另一条信号通路;③BMP-2与TGF-β1在骨折愈合过程中可能存在协同作用。 To investigate the expressions of transforming growth factor β1 (TGF-β1)and bone mor- phogenetic protein-2 (BMP-2)in human mandible fracture callus and their quantity changes in the process of healing. Method: Thirty callus samples from the fractured mandible bone stumps were collected during operation, and two callus samples were collected from the angle-square jaw patients as controls. The expressions of TGF-β and BMP-2 were test by the immunohistochemistry technic-SABC-staining in different periods of human fractured man- dibular callus and in osseous tissue of normal angle of mandible. Result:The TGF-β1 and BMP-2 were expressed in callus of different periods but not in normal bone tissue. The expression of TGF-β1 increased slowly during the first three weeks after fracture and reached its maximum in the third week, and then weakened gradually. The ex- pression of BMP-2 increased gradually during the first two weeks after fracture and reached its maximum in the second week, then the expression weakened gradually. Conclusion: ①BMP-2 may be one of the factors promoting the repair of fracture. ②TGF-β1 could be another signal pathway in repairment of fracture. ③There could exist some synergistic effects between TGF-β1 and BMP-2 in the process of fracture healing.
