摘要
为了研究不同深度土壤中微生物的多样性以及优势菌群,采集来自于不同深度的土壤样品,利用SDS苯酚/氯仿/异戊醇提取土壤中总DNA。扩增16SrDNA的V3可变区,变性梯度凝胶电泳进行分析。结果表明:土壤层自下向上各层香农指数分别为1.56、1.59、1.54、1.57、1.57、1.57、1.59、1.54、1.59、1.59、1.59。不同深度土壤层优势菌群数量和种类经测序分析并将结果在NCBI数据库比对,主要的优势菌属为Thermoanaerobacterales、Thermoanaerobacteraceae、Cyanobacteria、Firmicutes、Proteobacteria、Gammaproteobacteria、Oscillatoriophycideae、Clostridia、Deltaproteobacteria、Desulfobacterales。
This paper mainly studied the microbial diversity in soil of different depth and the advantage bacterium group. The samples were from 50 cm/45 cm/40 cm/35 cm/30 cm/25 cm/20 cm/15 cm/10 cm//5 cm/0 cm away the surface in the same place. Total DNa from soil were extracted using SDS phenol/chloroform/isoamyl alcohol extraction method. V3 variable region of 16 SrONA was amplified and was analysized using the denaturing gradient gel electrophoresis. The results showed that the Shannon index was 1.56,1.59,1.54,1.57,1.57,1.57,1.59, 1.54,1.59,1.59,1.59 of different soil clay. The number and variety of advantage bacterium group was different in different depths of soil layer, Sequencing analysis results were compared in the NCBI database and the main advan- tage bacterium were Thermoanaerobacterales , Thermoanaerobacteraceae , Cyanobacteria , Firmicutes , Proteobacteria ,Alphaproteobacteria , Oscillatoriophycideae , Clostridia , Deltaproteobacteria and Desulfobacterales.
