热应激对小鼠十二指肠组织形态和Caspase-3表达的影响

摘要 旨在阐明热应激对小鼠十二指肠形态结构的影响及其分子机制,8周龄ICR雄性小鼠,每天热处理2 h(环境温度42℃),连续12 d,分别在热处理第0、4、8和12天后,通过HE染色和免疫组织化学法观察小鼠十二指肠形态变化及Caspase-3蛋白的表达。结果显示,小鼠体表温度和直肠温度在热处理后显著升高(P<0.05)。随着热暴露天数的增加,小鼠十二指肠上皮细胞脱落,肠绒毛随着热处理天数增加而缩短,细胞凋亡蛋白Caspase-3表达随着热处理天数增加而增加。热处理引起小鼠热应激,热应激使小鼠的十二指肠形态结构受损,绒毛长度变短,细胞凋亡蛋白Caspase-3表达增加。 To investigate the effect of heat stress on the morphology of the duodenum and its molecular mechanism in mouse, eight-week-old mice were exposed to the elevated ambient temperature(42℃ )daily for 2 h. During the period of 12 consecutive days, mice were sacrificed on days 4, 8 and 12 immediately following heat exposure. The duodenum morphological changes were observed by HE staining and immunohistochemical detection of Caspase-3 expression in the duodenum. Heat-exposed mice presented significant increases in rectal and body surface temperature(P 〈0.05). With the increase of heat treatment days, the lengths of duodenal villus become shortened and epithelial cells loss. The Caspase-3 protein expression is elevated in the duodenal villus of heated-mice. These results suggest that heat stress damage to the duodenum morphology and increase the expression of apoptosis protein Caspase-3 in the mice.
