摘要
目的探讨螺旋藻多糖(PSP)对实验性胃溃疡大鼠的保护作用及其与前列腺素E2(PGE2)的关系。方法采用幽门结扎法制备大鼠胃溃疡模型,将50只SD大鼠随机分成正常组、模型组、奥美拉唑组(1.8 mg/kg)和高、低两个剂量的PSP治疗组(400 mg/kg和200 mg/kg),观察各组大鼠溃疡指数,测定其胃液分泌量,采用酸碱中和滴定法测定总酸度,麦特毛细管法测定胃蛋白酶活性,并用酶联免疫法检测大鼠胃组织PGE2的含量。结果螺旋藻多糖高剂量组(400 mg/kg)能降低大鼠胃溃疡程度,溃疡抑制率为41.67%,抑制胃液的分泌,降低胃液总酸度[(84.39±7.38)mmol/L]和胃蛋白酶活性[(178.36±55.02)U/mL],并可显著升高大鼠胃组织PGE2含量[(13.07±4.75)ng/mL],与模型组比较,差异有统计学意义(P<0.01)。结论螺旋藻多糖对实验性胃溃疡有促进溃疡愈合、保护胃黏膜的作用,这种作用可能与增加胃黏膜PGE2合成有关。
Objective To observe the protective effects of PSP on rats with gastric ulcer and the relationship between it and PGE2.Methods The gastric ulcer models were established by pyloric ligation.50 SD rats were randomly separated into five groups,including two groups of PSP (400 mg/kg of high dose and 200 mg/kg of low dose),Omeprazole group (1.8 mg/kg),model group,and normal group.The ulcer index,gastric juice volume were observed; total acidity was measured by neutralization titration method,the activity of pepsin was measured by capillary method,and euzymelinked immunosorbent assay was used to detect the PGE2 contents in tissues.Results The degree of gastric ulcer reduced,the ulcer inhibition rate was 41.67%,activity of pepsin [(178.36±55.02) U/mL] and gastric acid secretion [(84.39±7.38) mmol/L] reduced in the PSP group (400 mg/kg),and the contents of PGE2 [(13.07±4.75) ng/mL] increased remarkably,compared with the model group,the differences were statistically significant (P < 0.01).Conclusion PSP shows effects of promoting ulcer healing and protective effects on gastric mucosa,and its mechanism may be related to increasing of the composing of PGE2.
